Objective:This study examined the active ingredient luteolin of Bizhongxiao decoction on T lymphocyte proliferation in vitro,as well as NF-κB signal pathway to explore the anti-inflammatory mechanism of luteolin on treatment of rheumatoid arthritis.Methods:In order to explore the role of luteolin on immune regulation, we studied the effect of luteolin on proliferation and activation of T lymphocyte,and effect on NF-κB signal pathway in vitro.To stimulated T cell sources in mouse spleen lymphocytes with phorbol ester(PMA) plus ionomycin(ionomycin,Ion) with different final concentrations of luteolin co-culture with T cells,T lymphocytes are divided into the experimental groups:T cells Group(control),T cells+PMA+Ion Group,T cells+ PMA+Ion+10μM luteolin-group,T cells+PMA+Ion+20μM luteolin-group,T lymphoid Fine+PMA+Ion+30μM luteolin Group, total of five groups,determined by MTT cell proliferation;to detect the effect of luteolin on Ion plus PMA stimulated T cells of the expression of the phosphorylation of IκBαand P65 into the impact of nuclear by Western blot.Results:(1) MTT assay shows after 48 hours stimulated by PMA plus Ion,the proliferation of T cells increased obviously,luteolin(5-30μM) can inhibit the lymphocyte proliferation induced by PMA plus Ion in a dose dependent manner(P<0.01).(2) Western blotting assay shows after stimulated by PMA plus Ion for 30 minutes,the nuclear translocation of P65 is the most,luteolin(10μM, 20μM,30μM) blocked nuclear translocation of P65 in T cell in a does dependent manner;After stimulated by PMA plus Ion for 12 hours,the expression of phosphorylation of IκBαis the most,luteolin(10μM, 20μM,30μM) depressed the phosphorylation of IκBαstimulated by PMA plus Ion in a does dependent manner(P<0.01).Conlusion:(1)Luteolin could inhibit the lymphocyte activation and proliferation,it indicate that lueolin have anti-inflammatory effect.(2) Luteolin inhibits NF-κB signaling pathway of activated T cells, suggesting that luteolin can inhibit the function of T cell by inhibiting NF-κB signaling pathway,so as to achieve the effect of anti-inflammatory.
