Purpose:Experimental study of this item is based on the "Collateral", using appropriate, safe and effective dose in rabbits Huoxuetongluo improve revascularization in acute myocardial infarction myocardial no-reflow after the Organization of cytosolic phospholipase (cPLA2-y) geneexpression, from the molecular level the mechanisms of myocardial microcirculation and lead to coronary heart disease a preliminary study of myocardial protection provided the basis for clinical research data.Material and method:30 adult New Zealand white rabbits were randomly divided into 3 groups (n = 10), after ligation of left ventricular support after 90min,120min loose ligature infusion models of acute myocardial ischemia-reperfusion injury model; since the beginning of reperfusion,Ⅱgroup,Ⅲgroup were given normal saline from the ear vein (based upon 0.3ml·kg-1·min-1 rate of continuous infusion), Ginaton (initially 2mg·kg-1 intravenous injection, and thenby 0.3mg·kg-1·min-1 rate of continuous infusion).120min reperfusion when to stop infusion.Right room at the end of the experiment blood samples collected in serum creatine kinase (CK), lactate dehydrogenase (LHD) leakage rate, in the office after the right atrial appendage myocardium by reverse transcriptase polymerase chain reaction (RT-PCR)of myocardial tissue cPLA2-y gene expression, and the other, left ventricular myocardium by electron microscopy morphology of myocardial ultrastructure.Results:1. With the normal group, model group and Ginaton group of cPLA2 mRNA expression had significantly higher (P<0.01), Ginaton group of cPLA2 mRNA expression increased in the normal group, but with the model group was significantly lower thangroup (P<0.01).2. Compared with the sham group model group Myofibrillar fragmentation, dissolution, disappearance of a blank area within the Mi sarcoplasmic reticulum swelling, outer membrane appear Mi incomplete myelin degeneration, glycogen disappeared, intercalated disc swelling.Compared with model group, treatment group Ginaton myofibrillar sarcomeres aligned Z lines clearly marked, sarcoplasmic reticulum Mi have increased, glycogen particles increased, intercalated disk close to normal.3. Jinnaduoming significantly reduced serum CK, LDH activity.Conclusion:1.Ginaton improve myocardial ischemia reperfusion injury of cardiac systolic function, reduced cPLA2-γ gene expression, enzyme leakage, reduce the damage of myocardial ultrastructure.2. Ginaton on myocardial ischemia-reperfusion injury has a protective effect, its mechanism may be related to its inhibition of oxygen free radicals, inhibit lipid peroxidation effects.
