| Objective:By finding the difference of the extent of the damage for cardiac muscle which is affused by blood after being lack of blood, theoretical base can be built for the clinical prevention and treatment for ischemia- reperfusion damage.Methods:30 male rabbits are selected, which are 6~12 months old and weigh 2.5~3kg. These rabbits are divided into 6 groups, with each group 5 rabbits, and the models that cardiac muscle is lack of blood are built. The cardiac muscle tissues of the model are cut to make paraffin slices, then color the slices by HE and observe them. Then troponin I in muscle are detected. Annexin-V-Flous FCM are used to detect the indices related to cell Apoptosis. And the detection system of cell Apoptosis by DeadEnd colorimetry are used to detect the morphologic of cell Apoptosis Then transmission and electron microscope is used to observe the ultrastructure of the cardiac muscle tissues.Results:1.The morphologic observation of HE coloration of cardiac muscle tissues.the muscle cells of the lack-of-blood groups are swelling, marcoplasma is eosinophil coloration, the striped muscle is blurry or disappear,the nuleus is concentrated and inflammation cells sockage appears in the interstitial tissue. The cardiac muscle fibre is denaturalized, which shows the damage of cardiac muscle cells.2.the results of the detection of troponin I: the second group arrives at the peak value, the value of the last three groups in turn reduces more by the delayed time longer. 3.the detection results of cell Apoptosis: the detection results by the different methods show that the number of the Apoptosis cardiac muscle cells increases with the lack-of-blood time delaying. And comparing with the control group the difference is significant(P<0.05).4.The observation of ultrastructure shows that the intracell ultrastructure is damaged and the extent of the damage supports the detection results of troponin I.Conclusion:1.When the coronary artery has been interdicted 10 minutes to 60 minutes and then the cardiac muscle is affused, the cardiac muscle cell will be damaged. Among those when the coronary artery has been interdicted 10 minutes the damage is the biggest. With the time delayed longer, however, the extent of damage will be less. When the coronary artery has been interdicted more than 60 minutes the damage is the smallest.2.The damage from reperfusion can generate the Apoptosis procedure of cardiac muscle cell. When the coronary artery has been interdicted 10 minutes and then the cardiac muscle is affused, cell Apoptosis is observed. With the time delayed longer, the number of the Apoptosis cardiac muscle cells increases. When the coronary artery has been interdicted 10 minutes to 30 minutes and then the cardiac muscle is affused, the amplitude of Apoptosis rate is the largest. When the coronary artery has been interdicted 30 minutes to 60 minutes,the Apoptosis rate is still increasing, but the amplitude is less.3.The adaption in advance to absence of blood can lessen the extent of the damage and it can also decrease the Apoptosis rate of cardiac muscle cell.4.The extent of damage does not relate to the Apoptosis rate. |
