Purpose: To study the chemical constituents from the rhizome of Pulsatilla cernua (Thunb.) Bercht. et Opiz and especially for the separation of triterpenoid saponins and the structural determination of these compounds.Method: By the modern chromatography techniques (silica gel, macroporous resin, ODS and HPLC, etc), triterpenoid saponins were isolated from the rhizome of Pulsatilla cernua (Thunb.) Bercht. et Opiz. Their structures were elucidated by the spectroscopic analysis (1H NMR, 13C NMR, ESI-MS, HMQC and HMBC) and chemical methods.Result: Ten triterpenoid saponins were isolated from the rhizome of Pulsatilla cernua (Thunb.) Bercht. et Opiz. These compounds were identified as hederagonec acid (â… ); hederagenin (â…¡), hederagenin 3-O-α-L-arabinopyranoside (â…¢), hederagenin 3-O-β-D- glucopyranosyl-(1â†'2)-α-L-arabinopyranoside (â…£), 3-O-α-L-arabinopyranosyl hederagenin 28-O-α-L-rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranoside (â…¤) , 3- O-α-L-arabinopyranosyl bayogenin 28-O-α-L-rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl- (1â†'6)-β-D-glucopyranoside (â…¥) , 3-O-β-D-glucopyranosyl-(1â†'4)-α-L-arabinopyranosyl hederagenin 28-O-α-L-rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'6)-β-D-gluco- pyranoside (â…¦), hederagenin 3-O-β-D-glucopyranosyl-(1â†'4)-α-L-arabinopyranoside (â…§) , 3-O-β-D-glucopyranasyl-(1â†'2)-[β-D-glucopyranosyl-(1â†'4)]-α-L-arabinopyranosyl hedera- genin 28-O-α-L-rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranosyl ester (â…¨) , and 3-O-β-D-glucopyranosyl-(1â†'4)-α-L-arabinopyranosyl bayogenin 28-O-α-L- rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranosyl ester (â…©) .Conclusion: Compoundâ…©is a new triterpenoid saponin. Compoundâ…¥is first isolated from Pulsatilla cernua (Thunb.) Bercht. et Opiz. Compoundsâ… ,â…£,â…¤,â…¦,â…§, andâ…¨were obtained from this plant for the first time.
