| External antioxidant evaluation is an important method to evaluate antioxidants, due to the simple, quickly, low cost antioxidant detection system, the external evaluation method is widely used in food and medicine. The external evaluation systems are mostly based on pure chemical reaction system, reaction systems with different detection have different radical species, free radical method, types of free radical damage, damage detection theory, which lead to different methods of antioxidant testing results and poor correlation, even contradictory. Using different antioxidant testing methods to evaluate different antioxidants, the antioxidant capacity of antioxidants often vary greatly determined by the strength, thus it is difficult for the researchers to give a precise conclusion.Antioxidant screening test aimed at the human body that can be used to eliminate free radical antioxidants, for the purpose of prevention or treatment of disease, therefore, the external evaluation methods need to fit as much as possible to the internal methods to get the precise conclusion. Many external evaluation methods used are purely based on chemical reaction system, the free radical species and physiological free radical are inconsistent, the detection of free radical damage of external molecular species with material injury does not correspondent, therefore, the external evaluation methods often have little effect.We choose four common anti-oxidants as the test sample, using different methods of antioxidant testing to assess the effect of its antioxidant ability, make the correlation analysis of the evaluation results and classify them based on the results, to give the researchers a scientific basis for choose a right external evaluation method.Through the analysis of experimental results:First, the crystal violet, the 2 deoxy-D-ribose,TBA. The higher correlation between the three methods, so the capacity of antioxidant testing, the results obtained can represent hydroxyl radicals antioxidants scavenging or lipid protection. Second, Mitochondrial lipid peroxidation respectively with crystal violet, TBA, Inhibition of protein carbonyl formation, Hydrogen peroxide hemolysis has good correlation between the results of this study is focused on the recommended method. This method can be used to detect substances on biofilm system protection, or for lipid, protein protection. Third, remove DPPH radical, nitroso R salt by sperophotometry, t pyrogallol autoxidation system, the 3 methods is the result of a higher correlation. They can be used to detect anti-oxidant free radical scavenging ability of the majority. |
