Free radicals and antioxidants are frequently debated in the clinical and nutritional literature. A large amount of evidence have manifested that free radicals involved in many pathological activities including the processes of aging, cancer and other chronic diseases. Following the growing realization that a wide range of dietary supplements and antioxidant vitamins could be approved for preventive and therapeutic effects on chronic diseases due to their activities of scavenging radical and antioxidant. Sodium and potassium salts containing sulfur (SPSS) are important chemical reagents. They have been used in chemical, food, pharmaceutical, and medical industries. There are no data showing that sodium and potassium salts containing sulfur have antioxidant and free radical scavenging activities. In this study, we primary discuss the antioxidant and radical scavenging activity of the SPSS. In order to determine the antioxidant and radical scavenging activities of the SPSS, we use several different methods to evaluate the activity.Compatibility of commonly used antioxidants reasonablely can improve the antioxidant activity. To provide some experimental basis for selecting safe and effective combined antioxidants, we evaluated the combined and the individual antioxidant activities for three commonly used antioxidants-edaravone, GSH and vitamin C in this study. The main results were as follows:1. The activities of sodium and potassium salts containing sulfur as antioxidants and free radical scavengersObjective:We have investigated the antioxidant and free radical scavenging activities of SPSS by determining the total antioxidant activity, reductive activity, Fe2+ chelating activity, and superoxide-anion and hydroxyl radical scavenging activity in vitro; We also make a preliminary study on activities of the SPSS in vivo by measured SOD and MDA in mice serum.Method:Experiment in vrito:In this study, we have investigated the antioxidant and free radical scavenging activities of SPSS by determining the total antioxidant activity, reductive activity, Fe2+ chelating activity, and superoxide-anion and hydroxyl radical scavenging activity. To investigate the dynamic of the free radical scavenging abilities of SPSS, we evaluated the kinetics of the SPSS scavenging ABTS+ based on the kinetic spectrophotometric free radical processing measurement method in this study.Experiment in vivo:20-22g health male KM mices are randomly divided into Na2SO4, K2SO4, Na2SO3, K2SO3, Na2S2O3, K2S2O3, Na2S2O4, Na2S2O5, K2S2O5, Vitamin C, water-control and control group. To evluate the antioxidant and radical scavenging activities, we adopt the way of lavage for SPSS solution on the mice for a week in this study. Then we measured SOD and MDA in mice serum.Results:Experiment in vrito:Determine the antioxidant and free radical scavenging activities of SPSS in vitro:Our research indicates that the SPSS except Na2SO4 and K2SO4 have significant antioxidant and free radical scavenging features. The activities were associated with the valence of the sulfur. The antioxidant and radical scavenging activities of the S4+ were stronger than those of the others.Evaluate the dynamic of antioxidant and free radical scavenging activities of the SPSS in vitro:In this study, the SPSS except Na2SO4 and K2SO4, as free radicals scavengers, have a fast and stable dynamic character in vitro, which is necessary and helpful for further understand the SPSS’s antioxidant and scavenging free radicals properties.Experiment in vivo:1). The survival rate of mice:In this study, all mice of 12 group are remaining alive. Their diet, sleep and other activities have no significant change.2). Changes in body weight:The weight increment of the SPSS group were lower than that of water-control. The significant difference between Na2S2O3, Na2SO3, K2SO3, Na2S2O5, K2S2O5 group and water-control group were apparently.3). The change of blood glucose:The comparation between the experiment group and control was no significant difference. And the blood glucose of all group were kept in the normal range.4). The change of SOD:The SOD of SPSS except Na2SO4 and K2SO4 group were lower than that of control group. It explained that the amount of O2-in the blood has gone down. Therefore, it could indicate that the SPSS except Na2SO4 and K2SO4 have the ability to scavenge the oxygen radicals. On the other hand, this study could illustrate the SPSS except Na2SO4 and K2SO4 have antioxidant and free radicals scavenging activities.5). The change of MDA:The MDA of vitamin C group and SPSS group except Na2SO4, K2SO4 and Na2S2O4 were lower than that of control group. It indicated that the amount of lipid peroxide was induced. Likewise, it illustrated that the SPSS except Na2SO4, K2SO4 and Na2S2O4 have antioxidant and oxygen free radicals scavenging activities.Conclusion:Our research indicates that the SPSS except Na2SO4 and K2SO4 have significant antioxidant and free radical scavenging features in vitro and in vivo. This study indicate that they have a fast and stable dynamic character in vitro, which is necessary and helpful for further understand the SPSS’s antioxidant and scavenging free radicals properties. This study illustrate the SPSS at this dosage was not affect the diet、sleep and other activities of mice. As a consequence, the SPSS except Na2SO4 and K2SO4 have a stronger antioxidant and free radical scavenging activity.2. The comparative analysis of different combination antioxidants on their antioxidant capabilitiesObjective:In order to provide reference for clinical application and food additive, we evaluated the combined and the individual antioxidant activities for three commonly used antioxidants-edaravone, GSH and vitamin C.Methods:Comparing the multiple and the individual antioxidant activities for three antioxidants were carried out by the determination of total antioxidant activity assay, reductive activity assay, superoxide anion scavenging assay and hydroxyl radical scavenging assay in this study.Results and Conclusion:Results showed that the combined antioxidants acted synergistically in scavenging free radical, increased remarkably antioxidant activity at the same molar concentration. According to our date, the rationality of different antioxidant combination should be taken into consideration in the subsequent clinical application.
