Objective: To investigate the effect of ganoderma lucidum spores powder on NCAM-1 and NCAM-L1 of nerve cells in hippocamp of rat with epilepsy emblazed by PTZ and study the mechanism of epilepsy and improvement mechanism by ganoderma lucidum spores powder. Methods: 30 male Wister rats were divided into three groups randomly: ganoderma lucidum spores powder group; epilepsy model group; and blank control group, every group had 10 rats. The sub-eclampsia dosage of PTZ (35mg/kg) was used daily to make epilepsy model for 28 days; normal saline was given to blank control group by intraperitoneal injection with the equal capacity. Ganoderma lucidum spores powder group was given ganoderma lucidum spores powder (30g/L) from stomach pouring, the volume was 10.0ml/kg; normal saline was given to blank control group and epilepsy model group with equal volume. If emblazed standard come to levelâ…¡for 5 days, kill rats by decollation and get brains quickly at lower temperature to measure the NCAM-1 expression by RT-PCR,Western-blot and immunohistochemical staining; and to measure the NCAM-L1 by RT-PCR. Results: All the rats of ganoderma lucidum spores powder group and epilepsy model group achieved emblazed standard. In immunohistochemical staining experiment NCAM-1 positive cells of epilepsy model group increased obviously compared with blank control group(P<0.01),and ganoderma lucidum spores powder group decreased obviously compared with epilepsy model group (P<0.01). In western-blot experiment NCAM-1 expression of epilepsy model group increased obviously compared with blank control group (P<0.01), and ganoderma spores powder group decreased obviously compared with epilepsy model group(P<0.01). In RT-PCR experiment NCAM-1 and NCAM-L1 mRNA of epilepsy model group increased obviously compared with blank control group (P<0.01), and ganoderma spores powder group decreased obviously compared with epilepsy model group (P<0.01). Conclusions: Both of NCAM-1 and NCAM-L1 of nerve cells may have relation with the starting and developing of epilepsy caused by PTZ; Ganoderma lucidum spores powder could inhibit the growth of axons of neurons and transmembrane signal transduction in the nervous system, and reduce cell-cell adhesion through reducing over expression of NCAM-1 and NCAM-L1. Synaptic plasticity of the nervous system was reduced and epilepsy was improved. Ganoderma lucidum spores powder is an assistant medicine to cure epilepsy, and it may become clinical drug to protect brain agaist epilepsy.
