Objective:To observe and discuss the influence of Ganoderm a lucidum spores on the SS,NPY,5-HT and ATPase content in the brain of rat with epilepsy caused by Pentetrazole (PTZ)。futurely, study the occurring mechanism of epilepsy and operation mechanism of ganoderma lucidum spores powder. Methods:Thirty male Wister rats were divided into three groups randomly: ganoderma lucidum spores powder group; epilepsy model group; and blank contrast group, every group has ten rats. Use the sub-eclampsia dosage of PTZ (35mg/kg) daily to make epilepsy model for 28 days; normal saline was given to blank contrast group by intraperitoneal injection with the equal capacity. Ganoderma lucidum spores powder groupwas given ganoderma lucidum spores powder (30g/L) from stomach.pouring volume was 10.0ml/kg; normal saline was given to blank contrast group and epilepsy model group from stomach with the equal capacity. Register the spasm level and latent period and enduring time after injection of PTZ daily, emblazed standard was that levelⅡcontinued for 5 days. put rats to death by decollation, get brains quickly at lower temperature, measure the content change of NPY in cerebral cortex,hippocampus and plasma by radioimmunoassay; measure the immune reactivity of SS and 5-HT in hippocampus and cortex with immunohistochemical staining;measure the content of ATPase in cerebral cortex and hippocampus with chemical chromatometry method.Results:All the rats of ganoderma lucidum spores powder group and epilepsy model group achieved emblazed standard.The content of NPY in cortex and hippocampus increased obviously in the epileptic model group as compared with the blank control group,[(245.96±31.13)pg/ml,(270.02±29.32)pg/ml] ; [(185.47±16.28)pg/ml,(209.38±43.30)pg/ml](P<0.01),The content of NPY in plasma increased obviously in the epileptic model group as compared with the blank control group, [(1037.89±122.73)pg/ml];[(797.93±74,45)pg/ml(P<0.01).The content of SS in cortex and hippocampus were significantly increased in the epileptic model group as compared with the blank control group,[(10.25±2.05)cell/HP,(12.75±1.90) cell/HP]; [(2.34±0.74) cell/HP,(4.75±1.38)cell/HP] (P<0.01).The content of 5-HT in hippocampus were significantly decreased in the epileptic model group as compared with the blank control group,[(5.87±1.35) cell/HP];[(11.12±3.13) cell/HP] (P<0.01).The content of ATPase in cortex and hippocampus significantly decreased in the epileptic model group as compared with the blank control group, (P<0.01).The content of neuropeptide Y in cerebral cortex and hippocampus were significantly decreased in the the Ganoderma lucidum spores group as compared with the epileptic model, [(206.11±45.07)pg/ml,(230.88±28.46)pg/ml] (P<0.05).The content of NPY in plasma were significantly decreased in the the Ganoderma lucidum spores group as compared with the epileptic model, (907.49±79.39)pg/ml, (P<0.05);The content of SS in cortex and hippocampus were significantly decreased in the the Ganoderma lucidum spores group as compared with the epileptic model, [(6.37±0.91) cell/HP,(7.37±0.91) cell/HP] (P<0.01).The content of 5-HT in hippocampus were significantly increased in the the Ganoderma lucidum spores group as compared with the epileptic model, [(8.75±2.94) cell/HP,(5.87±1.35) cell/HP]( P<0.05).The content of ATPase in cortex and hippocampus significantly increased in the the Ganoderma lucidum spores group as compared with the epileptic model,( P<0.05)Conclusions:SS,NPY,5-HT and ATPase may have relation with the occurring and developing of vrialec-emblazed epilepsy; ganoderma lucidum spores powder can weaken the noxious injure for over expression of SS on centre nerve system of rat, restrain inflammatory reaction of nerve system, Also can reduce the content of NPY after epilepsy, reduce excitability of illed nerve cells,prevent the damages of epilepsy to nerve cells; what is more, ganoderma lucidum spores powder can increase expression of ATPase strongly, and facilitate the immune reactivity of 5-HT, thereby play a role in protect the nerve cells. Ganoderma lucidum spores powder is an assistant medicine to cure epilepsy, it can be very useful in clinic application.
