ObjectiveObserve the effects of Ganoderma lucidum spores oil: (1) on the tumor growth of H22 bearing mice, S180 bearing mice, SGC-7901 bearing nude mice; (2)on the immune function of H22 bearing mice; (3)on the toxic reaction brought by CTX chemotherapy and 60Co radiation therapy, To identify the anticancer effect of Ganoderma lucidum spores oil, To clarify the relationship between the anticancer effect and the immunoregulation effect of Ganoderma lucidum spores oil; In order to provide the pharmacological support of Ganoderma lucidum spores oil soft capsule for the new drug research and its supplication in clinic.Methods:1. The effect of Ganoderma lucidum spores oil on the tumor growth of tumor bearing mice: (1)establish the tumor bearing mice model of H22 and S180, observe the effect of Ganoderma lucidum spores oil and positive control drug on the tumor growth of H22 and S180 by decoherencing tumor and calculating inhibition ratio of the tumor weight. (2)establish the tumor bearing nude mice model of human gastric adenocarcinoma SGC-7901; observe the effect of the drugs on the tumor growth of SGC-7901 by measuring the tumor diameter regularly.2. The effect of Ganoderma lucidum spores oil on the immune function of H22-bearing mice: establish the tumor bearing mice model of H22, Study the effect of the drugs on the phagocytosis function of the endothelial system (RES)of the H22-bearing mice by carbon clearance test; study the effect of the drugs on the humoral immune function of H22-bearing mice by serum erythrocytolysin test; observe the effect of the drugs on delayed type hyper sensitivity reaction of H22-bearing mice by DTH test; observe the effect of the drugs on the cytokine of TNF-α,IFN-γin blood serum of H22 bearing mice by ELISA kit. 3. The effect of Ganoderma lucidum spores oil on the Toxic reaction brought by CTX chemotherapy and 60Co Radiotherapy: establish the tumor bearing mice model of H22; study the effect of the drugs on the toxic reaction of the chemotherapy and radiotherapy by detecting the number of the peripheral blood leucocyte, the number of bone marrow karyota; spleen index, thymus index of H22 bearing mice.Results:1. The inhibitory effect of Ganoderma lucidum spores oil on the tumor growth of tumor bearing mice: Ganoderma lucidum spores oil at high dose could significantly inhibit the H22 tumor, the inhibition ratio is 38.5%,42.7%,44.3%; Ganoderma lucidum spores oil at middle and high dose could significantly inhibit the S180 tumor, the inhibition ratio is 32.6%, 38.2%, 42.2% and 36.0%, 41.9%, 45.9%; Ganoderma lucidum spores oil at high dose could significantly inhibit the SGC-7901 tumor of the tumor bearing nude mice, the relative tumor growth rate T/C(%) is 59.1%, 59.2%,compared with negative control group, there is significant difference(P<0.05).2. The effect of Ganoderma lucidum spores oil on enhancing the immune function of H22-bearing mice: Ganoderma lucidum spores oil at each dose could significantly raise clearance index number K, phagocytic quotient a, and raise the swelling level of ear and the swelling rate of ear of the H22-bearing mice, compared with negative control group, there is significant difference (P1<0.05, P2<0.01); Ganoderma lucidum spores oil at high dose and middle dose could significantly enhance the serum erythrocytolysin level of H22-bearing mice, compared with negative control group, there is significant difference (P1<0.05, P2<0.01); Ganoderma lucidum spores oil at high dose could significantly raise the IFN-γand TNF-αlevel of blood serum of H22 bearing mice, compared with negative control group, there is significant difference (P<0.05).3. The effect of Ganoderma lucidum spores oil on Reducing Toxicity of the CTX chemotherapy and 60Co Radiotherapy: Ganoderma lucidum spores oil at high and middle dose could significantly relieve the decrease of the number of the peripheral blood leucocyte, the number of bone marrow karyota, spleen index and thymus index of tumor-bearing mice hurted by CTX at a big dose or 60Co, compared with negative control group, there is significant difference (P1<0.05,P2<0.01).Conclusions:1. Ganoderma lucidum spores oil could effectively inhibit the tumor growth of H22, S180 and SGC-7901 beared by mice, indicate Ganoderma lucidum spores oil as an effective anticancer drug can have wide clinical application perspective.2. Ganoderma lucidum spores oil could promote the cell mediated immune function, the humeral immune function and the level of cytokine related to tumor, indicate that Ganoderma lucidum spores oil has wide effects on immune system, this may be an significant way by which Ganoderma lucidum spores oil could effectively inhibit tumor growth.3. Ganoderma lucidum spores oil could decrease the toxic reacion of depauperating immune organ and depressing bone marrow, etc. brought by CTX chemotherapy and 60Co radiotherapy. Indicate that Ganoderma lucidum spores oil could be used in tumor therapy combined with chemotherapy and radiotherapy, and may have a synergia and attenuated clinic effect.
