Objective:To dynamic observe the distribution of the extracellular DNA in the formation of the candida albicans biofilm in the root canal; and through observed the influence of DNaseⅠto the formation of the candida albicans biofilm, study the function of the extracellular DNA in the formation of the candida albicans biofilm in the root canal. To find a new direction to overcome the problem of biofilms.Methods:Collected candida albicans from root canals, Biofilms were formed on 2.0-cm2 polymethylmethacrylate strips, the distribution of the extracellular DNA was observed by CLSM in different times. Added DNaseⅠto the candida albicans biofilm in six defferent times, To observe the change of the candida albicans biofilm picture by CLSM.At the same time, quantitative analysis the growth vigour of the biofilm befor and after the function of DNase1 by XTT reduction method. All figures will be measured for 6 times,compute the mean and standard deviation,analyse the figures by statistical software.Result:Candida albicans biofilm contain the extracellular DNA, and the extracellular DNA inceases with the development of the biofilm formed by candida albicans biofilms. The early stage of the candida albicans biofilm after the treatment of DNaseⅠ,only leave behind several cells .The mid-term stage of the candida albicans biofilm after the treatment of DNaseⅠ,only shaped a little slight bacterial colony. The maturation stage of the candida albicans biofilm after the treatment of DNaseⅠ,only shaped a loose dicty structure. XTT reduction method showed that after the treatment of DNaseⅠ, the growth vigour of the Candida albicans biofilm in experimental groups is reduced compared with control groups, especially the young candida albicans biofilms.Conclusion:The characteristics of the candida albicans biofilms is able to bear the rule medicinal, extracellular matrix of the biofilms consist of extracellular monosaccharide,extracellular protein,extracellular DNA and so on. The extracellular DNA relate to the collection of cells. candida albicans biofilm contain the extracellular DNA, and the extracellular DNA inceases with the development of the biofilm formed by candida albicans. DNaseⅠcan inhibit the development of Enterococcus faecalis biofilm grouped to any times,especially the young ones,it also can decompose mature biofilm,which all prove that extracellular DNA is significant in the formation and development in candida albicans biofilm.
