Objectives: To observe Candida albicans in vitro biofilm formation, structure, shape model, and show its strong drug resistance, and to study the role of the relevant drugs on the mechanism of biofilm, in order to explore the treatment of respiratory tract Candida albicans yeast infection of the feasibility of biofilm.Methods: Candida albicans in vitro biofilm formation was established in the study. The structure and model of biofilm was visualized by light microscopy and confocal laser scanning microscope. Scanning electron microscopy was used to observe Candida albicans in vivo biofilm model of rats. Candida albicans biofilm was intervene by the relevant drugs , then the quantity of hwp1 were analyzed by semi-quantitative RT-PCR. The metabolic activities and strong drug resistance of biofilms were measured using the XTT reduction assay .It is also used to understand the interference of Candida albicans biofilm by relevant drugs.Results:â‘ Candida albicans in vitro biofilm was established successfully. Confocal laser scanning microscope studies showed that yeast cells formed clumps along hypha-like aggregation. At last,the biofilm has a multi-layered film structure; Meanwhile, it was observed that aspirin did not inhibit the mycelial biofilm obviously, and fluconazole and ginger butter could inhibit biofilm formation of the mycelium.â‘¡It was proved that Candida albicans in vitro biofilm had strong drugs resistance. The biofilm was susceptible to all the concentration of aspirin and ginger butter. However, 1mmol/l aspirin has no significant inhibition for 48 hours in the growth of biofilm. The biofilm was susceptible to 10% ginger butter on the different time points in the growth of biofilm;â‘¢Scanning electron microscope studies observed that rats with the catheter biofilm hyphae were intertwined with the spatial structure of reticular formation;â‘£Aspirin had no obvious inhibitory to the protein hwp1, whereas fluconazole and ginger butter inhibited the protein hwp1 Obviously. It is consistent with the confocal microscope results.Conclusion: Respiratory tract Candida albicans biofilm has a strong drugs resistance. The morphology of rats in vivo Candida albicans biofilm is confirmed.It is possible that Aspirin, ginger butter may provide the experimental supports for the future treatment of fungal infection biofilm.
