PARTⅠEXPRESSION AND SIGNIFICANCE OF AUGMENTER OF LIVER REGENERATION IN RATS WITH UNILATERAL URETERAL OBSTRUCTIONObjective To investigate the expression of augmenter of liver regeneration (ALR) in rats with unilateral ureteral obstruction (UUO) and its relationship with the renal interstitial fibrosis. Methods 48 male SD rats were randomly devided into group A (sham operated) and group B(UUO); each group contain 24 rats. The expressions of (ALR) and transforming growth factor-β1(TGF-β1) were detected by immunohistochemisty on day 3,7,14,and 28 after established UUO model. Results In sham operated rats,ALR were mainly expressed in Henle's loops, distal tubules and collecting ducts located in the medulla.However, ALR expression was gradually increased not only in above region but also in cortical area in UUO rats. ALR expression didn't observe on glomeruli in both group rats.The protein expressions of ALR and TGF-β1 in tubule-interstitium increased on day 3 and reach the maximum on day 28. The renal ALR expression was significantly associated with the increased expression of TGF-β1 (r=0.951.P<0.001). Conclusion The expression of ALR in renal tissue of UUO rats is active and may be associated with the increased expression of TGF-β1 and renal interstitial fibrosis.PARTⅡTHE PRELIMINARY STUDY IN THE EFFECT OF RECOMBINATION RAT AUGMENTER OF LIVER REGENERATION ON KIDNEY IN RAT UUO MODELObjective To investigate the effect of exogenous recombinant augmenter of liver regeneration (rrALR) on the renal fibrosis of rat models induced by unilateral ureteral obstruction (UUO). Mothed 96 male SD rats were randomly divded into four group: group A(sham operated), group B(UUO model), group C(model+ rrALR), group D(model+ blank plamid);each group contain 24 SD rats. Rats were sacrificed at day 3,7,14,28. The histopathologic changes were observed by HE,Masson staining. Protein expression of ALR,TGF-β1 andⅠ,Ⅲcollagen were detected using immunohistochemisty and Western blot. The expression of TIMP-1 in Rat renal was tested using RT-PCR.Results Compared with sham operated rats, the renal histopathological injury were seirous in Rat UUO Model at day 3,7,14 and 28 (P<0.05), and the expression level of ALR, TGF-β1 andⅠ,Ⅲcollagen, TIMP-1 on kidney were significant increased(P<0.05). However, the levels of expression in group C were higher than group B and D(P<0.05).Conclusion rrALR may increase renal fibrosis in Rat UUO Model by increasing the expression of TGF-β1 and TIMP-1.
