The Arctium lappa L.is biennial plants which belong to the burdock genus of the composite,and also named eastern ocean panax or"dalizi"and so on.The burdock roots has a great nutrition value and broad pharmacological activity,not only can be eaten but also as drug.This paper study the biological activity,extract and purify the oligosaccharide BOS from the burdocks roots.Using of separation and purification technology,we extracted and purified of sugars from the burdocks roots and had preliminary identification and optimization of the extraction condition.The oligosaccharides had been identified as a high purity single sugars.The best extraction conditions was the temperature 80℃,the solid-liquid ratio 1:10,time 60min,extracted twice.Using phenol - sulfuric acid method,the oligosaccharide content of BOS was measured for 96.2%.Mice were given Burdock Oligosaccharide with oral administration in advance for two weeks,and then administered 50%ethanol at the same time for a week.The model group animals body weight lost,liver index and serum ALT increased,the content of liver MDA,GSH and TG increased,GPX activity decreased,liver tissue pathological structural changes significantly,which all proved that the model of alcohol-induced liver injury in mice was established successfully.Burdock Oligosaccharide was found to have a significant reduction in body weight of the mice and have a promotion for the increase of serum ALT and liver GSH,and have a inhibition on the reduction of GPX activity and increase of TG and MDA content,and be able to significantly reduce the pathological changes caused by the alcohol in liver tissue.Mice were given Burdock Oligosaccharide with oral administration in advance for a week,and then were given carbon tetrachloride oil solution to make a model of liver injury.The content of serum ALT and AST were significantly increased in model group,content of liver MDA and GSH increased significantly,GPX activity was significantly reduced,liver tissue pathological structural changes significantly,which all proved that the model of CCL4 liver injury in mice was established successfully.Burdock Oligosaccharide was found to have a promotion for the increase of serum ALT and liver GSH,and have a inhibition on the reduction of GPX activity and increase of MDA content,but not be able to significantly reduce the pathological changes caused by the CCL4 in liver tissue.Mice were given Burdock Oligosaccharide with oral administration in advance for a week.We found that Burdock oligosaccharide could be effective in the promotion of mouse intestinal Lactobacillus and Bifidobacterium proliferation,and with the higher dose of oligosaccharide,the proliferation effect was more obvious,but the number of Enterobacteriaceae could be not impacted significantly.In conclusion,we found that Burdock oligosaccharide showed not only favorable but also negative side for mice.The mechanism would have to be further studied.
