| Objective:Study the therapeutical effect and mechanism of Okam on asthmatic mouse.methods:40 mouse were randomly divided into five groups,there are 8 mouse in each group,negative control(A),positive control(B),low dose Okam group(C),large okam Decoction group(D),Pulmicort group(E).The mouse models of asthma were made by the egg albumen sensitized method and were treated with low dose,large dose okam and Pulmicort for four weeks.Lung pathology were used to as indicator of airway inflammation and the expression of Matrix Metalloproteinases-9,Matrix Metalloproteinases -2,Tissue Inhibitor Metalloproteinase -1 and Tissue Inhibitor Metalloproteinase-2 in lung tissue were examined with immunohistochemical method.Results:â‘ Gasp symptom and inflammation in lung tissue in C,D,E group were.significantly lessen than B group, A group is normal.â‘¡Matrix Metalloproteinases-9,Tissue Inhibitor Metalloproteinase-1 and Matrix Metalloproteinases-9/Tissue Inhibitor Metalloproteinase-1 in A,C,D,E group were evidently less than those of B group.There is significant different between A,E group and C,D group.All of the parameters had no difference between A,E and C,Dâ‘¢Matrix Metalloproteinases-2,Tissue Inhibitor Metalloproteinase-2 and Matrix Metalloproteinases -2/Tissue Inhibitor Metalloproteinase -2 in A,C,D,E group were evidently less than those or B group.There is significant different between A,E group and C,D group.All of the parameters had no difference between A,E and C,D.Conclusion:Okam could treat asthma mouse,the possible mechanism is that Okam could down-regulate Matrix Metalloproteinases-9,Matrix Metalloproteinases-2,Tissue Inhibitor Metalloproteinase-1 and Tissue Inhibitor Metalloproteinase-2,so it regulate the balance between Metalloproteinases -9 and Tissue Inhibitor Metalloproteinase-1,Matrix Metalloproteinases-2 and Tissue Inhibitor Metalloproteinase-2.okam may be a new drug for the treatment of asthma. |
