ObjectiveTo investigate the effect of Okam on airway inflammation and remodeling in asthmaticmice which induced by ovalbumin.Methods32 mice of KUNMING STRAIN were divied into four groups randomly: Positivecontrol group, glucocorticoid inhalation group, Okam group and Negative group. Theasthmatic mice model were made by combining sensitization including injecting andaerosol inhalating of ovalbumin. The Positive controls received normal saline(3ml)inhalation daily during aerosol inhalating period. The Glucocorticoid group receivedbudesonide(0.4ml) and normal saline(3.6ml) inhalation before the daily OVA inspiration.The Okam group were orally treated with Okam daily (50mg/kg), and the negative controlswere given normal saline in sensitization and inhalation only. All mice were sacrificedbetween 24~48 houres after 42 days of inhalation and gavage. The expression ofinterferon-γ(IFN-γ),interleukin-4(IL-4) in broncho- alveolar lavage fluid (BALF) wereassessed by Enzyme-Linked Immunosorbent Assay(ELISA). The histological changes ofthe broncho-alveolar were evaluated after HE stain, and the expression of matrixmetalloproteinase-9(MMP-9) was determined by immunohisto-chemistry as well as thetissue inhibitor of metalloproteinase-1(TIMP-1).ResultsCompared with the Positive group, the total cell number and interleukin-4 level inBALF, MMP-9,TIMP-1 expression and the score of the broncho-alveolar tissue in Okamgroup were decreased greatly, and the interferon-γlevel increased markedly(p<0.01). Atthe same time, these indicators were also significant(p<0.05) compared with that of theNegative group. The total cell number,interleukin-4 and the interferon-γlevel in BALF,MMP-9,TIMP-1 expression and the score of the broncho-alveolar have no significantchanges between the Glucocorticoid group and the Okam group(t=0.19-1.501,p>0.05).ConclusionOkam may alleviate the inflammation of the bronchia and degrade the possibility ofairway remodeling.
