Objective:This study aims to observe the expression of interleukin-21(IL-21), thymic stromal lymphopoietin(TSLP), α-smooth muscle actin(α-SMA), type I collagen(Col-I)in lung tissue of mice and asthmatic mice,Discussion the possible immune mechanism of okma intervention therapy, which may be provide new theoretical basis in the treatment of asthma. Methods: 40 female BALB/c mice in Kunming strain by statistical method were divided into 4 groups: asthma model group, budesonide treated group, okma treated group and the control group, were marked group A、B、C、D. The mice models were established by Intraperitoneal injection of ovalbumin(OVA) and atomization OVA. B group were given Pulmicor(Budesonide 200ug), C group were given Okam(50mg/kg) by gavage, D group were received normal saline at all stsges. All the mice was executed between 1-2days after intervention ended. The bronchoalveolar lavage fluid(BALF) and the lung tissu were separately collected to check. Pathological changes of lung tissue in mice by hematoxylin-eosin(HE) staining to assessed, The levels of IL-21, TSLP,α-SMA, Col-I cytokines in BALF were tested by Enzyme-linked immunosorbent assay(ELISA), The expression level of IL-21, TSLP, α-SMA, Col-I by Immunohisto chemistry(IHC) tested.Result:This asthmatic model were established succeeded which all mice survived, The asthma symptoms and the pathological changes of lung tissue HE dyed are similar between the group B and the group C, both groups ease the group A, and serious the group D. The level of IL-21, TSLP, α-SMA, Col-I in BALF by ELISA tested and the expression level of IL-21, TSLP, α-SMA, Col-I in lung tissue by IHC tested between the group B and the group C, which have no obvious difference(P>0.05), While the two groups compared with the group A, Which were obviously decreased((P<0.05),While the two groups compared with the group D, Which were obviously increased((P<0.05).Conclusion:(1)The higher level expression of IL-21, TSLP,α-SMA, Col-I in BALF and lung tissue of asthma model mice groups, which may lead to the airway remodeling in asthmatic mice.(2)The level of IL-21, TSLP, α-SMA, Col-I in BALF and lung tissue of asthmatic mice reduced by okma and budesonide intervention, which reduced the expression of inflammatory mediators, preventing or reversing airway remodeling of asthmatic mice, and have both effect.(3)Okma can be used in the treatment of asthma in the future.
