Objective To study the expression of insulin-like growthfactor-1 (IGF-1),insulin-like growth factor-1 receptor(IGF-1R) in endometrial adenocarcinoma and the clinical significance.Methods The tissue specimens of 46 patients with endometrial adenocarcinoma were formalin-fixed,paraffin-embedded,cut and mounted onto the slides. Sections were stained for IGF-1,IGF-1R imrnunohistochemically.Twenty specimens of normal endometrium served as control.Results (1) percents specimens of the control group were positive staining ofIGF-1,without over expression.Positive expression of IGF-1 was in 60.87 % of the cancer,with over expression in 23.91% (moderate staining in 17 cases and strong staining in 11 cases). No significant difference of positive expression rate was observed between cancer and normal groups (X~2=0.004,P=0.947),but significant difference of over expression rates was obtained between normal and cancer groups (X~2=4.147,P=0.003). Positive expression and over expression of IGF-1 were not associated with FIGO stage (X~21=5.434,X~22=4.328;P1=0.066,P2=0.118),but over expression of IGF-1 correlated with cellular differentiation(X~2=7.163,P=0.028).(2) IGF-1R are membrane receptors. Seventy percents specimens of the control group were positive staining of IGF-1R,without over expression. Positive expression of IGF-1R was in 60.87% of the cancer,with over expression in 36.96% (moderate staining in 11 cases and strong staining in 17 cases).No significant difference of positive expression rate was observed between cancer and normal groups (X~2=0.511,P=0.475),but significant difference of over expression rates was obtained between normal and cancer groups (X~2=8.117,P=0.004). Positive expression and over expression of IGF-1R were not associated with the FIGO stage. (X~21=5.550,X~22=4.369;P1=0.062,P2=0.111) ,but over expression of IGF-1 correlate with cellular differentiation (X~2=8.584,P=0.014).Conclusions IGF-1 and IGF-1R are related to poor differentiation of the cancer. They may be used as an indicator for prognosis of the endometrial adenocarcinoma and provide some theoretic basis of the multiple strategies for management of the endometrial adenocarcinoma.
