Objective:The objective of this dissertation is to investigate the relationship between menstrual depression and high-estrogenic, by introducing experimental rats into high oestrogenic state using Estradiol Benzoate. Further, Jingqing capsules-a traditional Chinese medicine that disperses the stagnated Liver-Qi and regulates menstruation are introduced into the rat model, and their intervention mechanisms are investigated. Methods:We used young animal model of three to four months old impregnated SD female rats and randomly divided the rats into eight different experimental groups. Specifically speaking, they are the control group, the reserpine group, the Estradiol group, the Estradiol with sertroline group, the Estradiol with methyl testosterone group, the Estradiol with clomiphene group, the Estradiol with medroxy progesterone group and the Estradiol with Jingqing Capusule group. We performed vaginal smear test for each animal, and set up the files for its estrous cycle, then made the model during the estrous period. We injected Estradiol Benzoate into experimental rats on three consecutive mornings to introduce high oestrogenic state, and observed rats' behavioral changes as well as changes in their central neuro transmitter. We then compared the results with the classic depression model to evaluate the degree of success of oestrogenic modelling. At the same time of modeling, the above mentioned medicines were given for each group. The corresponding changes in vaginal keratinized cell index, behavior, serum E2 and P level and the ratio of E2/P, the quantity of 5-HT and DA of central neuro transmitter, as well as hippocampus- the neuron growth factor (NGF) and its mKNA expression were recorded and analyzed. Results:Our experiments demonstrate that Estradiol Benzoate can increase vaginal kerratinized cell index, increase serum E2 contents, decrease Brain stem neur- otransmitter 5-HT and DA concentrations and change the behavior of the animals: in open field test of the rats, the vertical motion, level motion and frequency of cleaning are decreased.
