PurposeTo observe the morphologic changes and the activity of c-fos,c-jun,NF-κB and Caspase-3 in rats retina after exposure to the operating microscope light,and study the protective effect and mechanism of Puerarin against retinal damage by light.。MethodsThe experiments were performed on fouty—eight healthy Sprague-Dawley rats which were devided into four groups—normal group,model group,low-dose group and high-dose group.All the groups except the normal one were continually exposed to the light of the operation microscope with an illuminace level of 22000±1000 lux for 30 minutes.The treated groups were injected daily Pue for one week after the light exposure.Then the pathological changes of retina were examined by light microscopy;and c-fos,c-jun,NF-κB and Caspase-3 into retina were tested.ResultsAfter the light exposure,the outer nucleus layer of retina in the model group became thinner and the regular arrangement of outer and inner segments were disordered,dividing line wasn't distinct.In the low-dose group the outer nucleus layer of retina got thicker than that of the model group.Pue can adjust and control the gene expression in phototonus of retina.ConclusionsPue could release the photic injury of retina,accelerate the renovation of retina after light exposure and ameliorate the function of retina.The mechanism might relate to control the expression of c-fos,NF-κB and Caspase-3 in phototonus of retina. |
