Objective To observe the expression of nuclear factor-KB(NF-KB), redox factor 1( Ref-1) and proliferating cell nuclear antigen( PCNA) in rat retina with ischemia-reperfusion injury treated with bFGF, then discuss the therapeutical mechanism of bFGF in the rtina ischemia-reperfusion injury.Method 24 Wistar rats were divided randomly into 8 groups. One was marked as normal group. The rest were marked as 0 hour (Oh),4h,8h, 12h, 24h, 48h and 72h group respectively, and in these groups, to the same rat, the right vitreous cavity was injected with bFGF as treated group, while the left was injected with Ringer's solution as ischemia group. To induce retinal ischemia, raising intraocular pressure was indicated, and 1 hour later stopping the pressure to recover the normal blood stream, which led to retinal ischemia-reperfusion injury. We detected the expression of NF-RB, Ref-1 and PCNA in inner retina with the SABC of immnohistochemistry by counting postive cells on high-power objective, then to analyse data with statistics.Results The expression of NF-K B and PCNA was negative in the normal rat retina. However NF-kB positive cells appeared a few in inner retina after Oh in ischemia-reperfusion injury, and more were detected in 4h to 12h , peaked at 12h to 24h and declined at 24h to 72h, and the expression level was still high. But the positiveexpression level in treated group were higher than those in the ischemia group in the same period. The expression of PCNA was also not detected in Oh in ischemia group, but it appeared and began to increase after 4h, peaked at 24h, then delined gradually , and at 48h to 72h it still appeared. The treated group was similar to the ischemia group, but the positive cells were much more. For Ref-1, it appeared a lot in retinal inner layer and a few in outer nuclear layer(ONL). It was no change on Oh to 4h in inner retinal layer in ischemia group and no expression after 8h in ONL, meanwhile decreased obviously in inner retinal layer, and no expression after 12h.The expression of Ref-1 in treated group was similar to ischemia group at Oh to 4h(p>0.05), but increased dramatically after 8h(p<0.05). Conclousion 1. No NF-K B and PCNA were detected in normal retina,but they increased gradually and had their peak time respectively in the process of ischemia-reperfusion injury. It may be a protective reaction. 2. Ref-1 exists in normal retina , decreases with ischemia-reperfusion.It plays a role of negtive regulation in retinal cells, and may be a kind of proteinase in the repair process of DNA damage. 3. bFGF can improve the activation of NF- K B, up-regulate Ref-1 and induce the expression of PCNA . In the process bFGF plays a role of anti-apoptosis, suggesting it has a therapeutical role in the retina ischemia-reperfusion injury.Postgraduate:Zhao Lux in(Ophthalmology)Directed by Prof. Niu Yingjun...
