Abstract/Summary:
Objective: Through the detection of the expression level of ADAMTS-4 in pulpy nucleus of human degenerated lumbar intervertebral discs, to investigate the relationship of ADAMTS-4 in the development of Lumber Disc Herniation(LDH),and to understand the pathology of Lumber Disc Herniation(LDH).This may provide an experimented proof for the diagnosis of clinical Lumber Disc Herniation(LDH).Methods:1.The fresh pulpy nucleus tissue was obtained surgically and were fixed in 10%formalin , imbedded in Paraffin and sliced by 4μm.2.The degeneration of pulpy nucleus tissue was observed with HE staining.3. The protein expression of ADAMTS-4 in experimental group (40 fresh pulpy nucleus of patients with lumbar disc herniation, 27 males and 13 females) and control group (8 fresh pulpy nucleus of patients with idiopathic scoliosis, 3 males and 5 females) were detected with immunohistochemical techniques lumber spine CT were examined at the same time.4.The results were analysed with SPSS 11.5 for windows.Results:1.Histopathologic examination of herniated intervertebral disc tissues showed granulation tissue at the peripheral portions of herniated discs.2. In normal annulus fibrosus,the express of the antibody of the ADAMTS-4 is very little .It can be found only in the nucleuses of chondrocyte.In degenerative lumbar intervertabral disc,the express is increased .The light density of ADAMTS-4 in degenerative lumbar intervertabral disc is markedly increased than the normal lumbar intervertabral disc(P<0.05).3.There is correlation between the content of ADAMTS-4 and the hernia size(P>0.05).Conclusions:1.The quantity of ADAMTS-4 increased in degenerative lumbar intervertabral disc. ADAMTS-4 involvement in herniated disc regression.2. There is no correlation between the content of ADAMTS-4 and the hernia size.
