Objective:This research investigated the expression and the relationship of ADAMTS-4and IL-6in degenerative intervertebral disc nucleus pulposus tissues, and then explore the effects of lumbar intervertebral disc degeneration.Methods:The degeneration group:nucleus pulposus degeneration of lumbar intervertebral disc, during the period of2012June to2013January in this hospital collecting the nucleus pulposus specimens of patients with prolapse of lumbar intervertebral disc, total of32cases (male19cases, female13cases),32years old-68years old, average (42.58±7.16) years old.The normal group:the same period in our hospital collecting the lumbar nucleus pulposus specimens of patients after lumbar traumatic operation, total of8cases (male6cases, female2cases),32years old-57years old, mean age (45.67±1.54).All specimens were made for2copies, one copy were fixed with PBS and became homogenate, Elisa was used to detect the content between degeneration group and the normal group of ADAMTS-4and IL-6; another tissues were paraffin-embedded and frozen, immunohistochemistry study was uesd on detecting the expression of ADAMTS-4and IL-6in the degeneration group and control group.Results:1.Elisa measurement of IL-6, ADAMTS-4degeneration group was significantly higher than normal subjects, measurement data were compared with two sample T test, the results is P<0.05, confirmed that it was statistically significant difference among groups.2.Immunohistochemistry S-P method was used to detect the express of intervertebral disc cells of ADAMTS-4and IL-6, in the normal group, the majority of samples ADAMTS-4and IL-6positive cell rate is low; in the degeneration group, at the same time, samples ADAMTS-5and IL-6positive rate are higher, results show as (-x) s, count data mining groups was analyzed by x2test (P <0.05) confirmed that difference was statistically significant;3. Combine with the relevant literature, using bivariate correlation analysis the content of IL-6and ADAMTS-4in the degeneration group, suggesting that the two correlation.Conclusion:Proteolytic hydrolase ADAMTS-4, interleukin-6in degenerative intervertebral disc was increased, and the two presented correlated, suggesting they may be involved in the degradation of proteoglycan, causing the intervertebral disc extracellular matrix loss balance, resulting in the degeneration of intervertebral disc.Through the study of IL-6, ADAMTS-4, provide a reference for future treatment on the level of molecular biology for intervertebral disc degeneration.
