Objectives: The research is aimed to proof-test whether the ingredient of withhold clysis of five-colours pulvis can successfully reach prostate and the research of it's pharmacokinetics.Methodologies: In the experimental research, the researcher clystered 72 mice with Five-Colours pulvis that was 31.2 times as much as that of the normal dose to an adult and got the prostate and blood at the following time point after the clysis respectively of 72 mice which was divided into 12 parts, each part had 6 mice: 5min,15min,30min, 45min, 60min, 75min,90min, 105min,120min,150min, 180min and 360min. Use the borneol in five-colours pulvis as inspect object and naphthaline as the internal standard. Dissect mice's prostates out equally and extract the borneol, and so do the blood. Detect it with Agilent 6890N gas chromatogram workstation with Agilent 7694E Headspace Sampler. Then compute the data with Excel2003,sas and 3p97 software.Results: It is found that borneol can be detected in the extract using gas chromatogram workstation. The concentration of borneol in prostate will be at peak value 90 minutes after the clysis, and the borneol in the blood is about 180minutes. Rectal clysis administration, Plasma concentration time curves of QLS fitted to a one compartment open model. One compartment model parameters: t1/2Ka75.90min,t(peak) 184.38min,Cmax165.58μg·ml-1,Lag Time4.29min,t1/2Ke240.07min,Ke0.0029min-1,Ka0.0091min-1,AUC97664.36μg·min,CL/Fs0.0026L·min-1。Conclusions: With the ability of reaching to the prostate tissue,borneol in prostate need more time to eliminate than blood.There are other paths to reach the prostate other than blood after clysis. Rectal administration, Plasma concentration time curves of QLS fitted to a one compartment open model.
