Objectives: The research is aimed to proof-test whether the ingredient of withhold clysis of five-colours pulvis can successfully reach prostate as well as the spermatophore tissues and do the research of it's pharmacokinetics. And, discuss the rationality of withhold clysis and its standardization in the clinical use today.Methodologies: In the general introduction of the bibliography, the essay points out the impracticability of the existing clinical withhold clysis on the basis of induction and deduction of 72 academic essays shown on domestic publications in the recent decade on withhold clysis treatment of chronic prostatitis and puts forward possible solutions to the standardization of withhold clysis in terms of mode of clysis, the depth of pipe in the anus, the volume of clysis liquid, the temperature of clysis liquid and the withhold time. In the experimental research, the researcher clystered 32 mice with Five-Colours pulvis that was 11.2 times as much as that of the normal dose to an adult and got the prostate and spermatophore at the following time point after the clysis respectively of 14 mice which was divided into 7 parts, each part had 2 mice: 30min, 45min, 60min, 90min, 120min, 240min and 360min, and got the prostate and its'blood at the following time point after the clysis respectively of the other 18 mice which was divided into 9 parts, each part had 2 mice:5min, 15min, 30min, 45min, 60min, 90min, 120min, 150min, 180min. Use the borneol in five-colours pulvis as inspect object and dodecane as the internal standard. Dissect mice's prostates out and spermatophore equally and extract the borneol, and so do the blood. Concentrate the extracts to 50μl and detect it with Agilent 6890N gas chromatogramwork station with Agilent 7694E Headspace Sampler. Then compute the data with Excel2003 and 3p97 software.Results: The research shows the present withhold clysis in clinical use is unreasonable in terms of mode of clysis, the depth of pipe in the anus, the temperature of clysis liquid and the withhold time. And the author's suggestion of standardization of mode of clysis, the depth of pipe in the anus, the temperature of clysis liquid and the withhold time are as fellows: clean the intestines before the clysis and use the side decubitus. 1. mode of clysis: Slow dropping (such as 40 drops per second). 2. the depth of pipe in the anus: 20-30cm in the anus. 3. the volume of clysis liquid: 100~150ml. 4. the temperature of clysis liquid: 42℃±0.5℃. 5. the preserving time: The longer, the better. The least is 2 hours.It is found that borneol can be detected in the extract using gas chromatogramwork station. The concentration of borneol in prostate and spermatophore will be at peak value 60 minutes after the clysis, and the borneol in the blood is about 90minutes. Rectal administration, Plasma concentration time curves of QLS fitted to a one compartment open model. One compartment model parameters: t1/2Ka47.10min,t(peak) 95.58min,Cmax5.15μg·ml-1,Lag Time23.35min,t1/2Ke97.34min,Ke0.0712min-1,Ka0.0147min-1,AUC1428.32μg·min,CL/Fs0.063L·min-1。Conclusions: With the ability of reaching to the prostate as well as the spermatophore tissue,blood, the concentration of borneol in prostate and spermatophore can reach the peak value about 60 minutes after the clysis, and in blood is about 90min. The content of borneol in target organ is more than in 1ml blood. Rectal administration, Plasma concentration time curves of QLS fitted to a one compartment open model. The unreasonable clinical withhold clysis treatment on chronic prostatitis need standardizing.
