| OBJECTIVE:Rheumatoid Arthritis(RA)is one of serious chronic diseases which threat public healthy in the world.Affecting sufferer's normal life quality and even can cause disability.Which belongs to Bi syndrome in TCM,it is also called "wan bi" or "kuang bi" because of it's intractability.Until now there is still no drug with curative effect and little toxicity and side effect.Therefore,it is very important to explore a comprehensive therapeutic plan which can ameliorate the symptom and prognosis of RA and decrease the adverse reactions.Animal models were established by injection of FCA for rat adjuvant arthritis.This research carries on a deep study on anti-inflammation,analgesic and hemorheology.and then to investigate the possible mechanism of jiu bai chui decoctionon RA.METHODS:(1)animals:healthy wistar rats 50,male,weighed 180～220g.kunming healthy mouse 80,half males and half females,weighed 18～22g.Ⅱclean grade.All of the animals were provided by Experimental Animal Center of medical college of Hubei Institute for Nationalities.(2)model duplication:BCG 50 mg inactivated in water of 70℃,mixed with the Liquid Paraffin and lanoline,then FCA are made successfully.Inject the healthy wistar rat' s right hindfoot with FCA 0.1ml respectively to duplicate AA rats model.(3)groupings and administrations:40 rats which are successfully duplicated are divided into 4 groups at random,and another group are not duplicated.That is A:normal control group B:blank model group C:zhengqingfengtongning control group D:treating group with high dose ofjiu bai chui decoction E:treating group with low dose of jiu bai chui decoction.While the normal control group and blank model group were treated with normal saline 10ml/kg·d,the rest 3 groups are treated with zhengqingfengtongning(30mg/kg·d),high dose of jiu bai chui decoction (6.62g/kg·d)and low dose ofjiu bai chui decoction(3.31g/kg·d)respectively.once per day,for 22 days.The first experiment mouse ear swelling induced by dimethyl benzene.The second experiment rats AA primary and secondary disease induced by CFAThe third experiment effect ofjiu bai chui decoction on Hemorheology in AA RatsThe fourth experiment the algesia caused by hot plate in the mice(4)Statistics:all data are showed with x±s,The t test was adopted in the intergroup comparison.RESULTS:(1)Analysis and evaluation of the animal model:according to the appearance,weight,activity,pathological changes seen by naked eyes and changes all over the body. we can arrive at a conclusion:the animal model is successful,which has been universally acknowledged.Synovitis,the hyperplasia of synovium,pannus formation and the destruction of cartilage and bone are the main pathological properties of RA.(2)both treating group with high dose ofjiu bai chui decoction and treating group with low dose of jiu bai chui decoction can abate the rat foot swelling,compared with the blank model group,which has significant difference(p＜0.05 or p＜0.01).compared with zhengqingfengtongning group,which also has difference(p＜0.05).The results show that The effect of the treating group with high dose of jiu bai chui decoction was superior to the zhengqingfengtongning group significantly,the treating group with low dose of jiu bai chui decoction has similar efficacy to the zhengqingfengtongning group(3)Index of Hemorheology displays that the treating group have many aspects' influence on AA rat Hemorheology,which were as follows:reduce hematocrit and plasma viscosity,as well as erythrocyte aggregation index,and then take on the decline of whole blood viscosity and total blood reductive viscosity.It suggested that the treating groups can improve the state of blood high coagulant and high viscosity, recover the normal state of microcirculation,increase the nutrition of limbs,alleviate the injuries of limbs.,through this methods,it has comprehensive effect on RA. (4)affect of inflammtion induced by dimethyl benzene and pain caused by hot-plate:jiu bai chui decoction can inhibit the acute exudative inflammation induced by dimethyl benzene obviously,compared with the normal saline group,which has significant difference(p＜0.05 Or p＜0.01);The effect of pain threshold of mouse was tested by hot-plate test,which shows that jiu bai chui decoction can notably improve the threshold of pain of the mouse caused by hot plank and prolonged the latent period of pain-threshold obviously,compared with the normal saline group,which has significant difference(p＜0.05 or p＜0.01).CONCLUSIONS:Experimental study proves that jiu bai chui decoction has remarkable anti-inflammatory and analgesic effects,which can inhibit the foot swelling of AA rats and murrine polyarthritis,raise rats' threshold of pain,improve the index of immune organ,reduce synovitis and synovial hyperplasia of diseased joints,prevent the destruction of arthrodial cartilage and bone;it also can improve the state of blood high coagulant and high viscosity,recover the normal state of microcirculation.The formulation of the prescription is reasonable,according with the pathogenesis of RA,definite therapeutic effect,which deserve further research.the study results provide the scientific basis for the clinical application and the clinical expansion. |
