The paper consists two parts:Literature review and experimental study.First Section:Literature SummaryThe Literature review introduces the experimental strvey of the acupuncture and moxibustion on the SCI.It contains three major portions about the acute spinal cord injury:the choice of the SCI model,the internal principle study of the acute spinal cord injury and the theropy of the acute spinal cord injury.Second Section:Experimental ResearchObjective:To discuss the protective mechanism of electroacupuncture(EA)in the acute spinal cord injury.Methods:Rats were divided into four groups stochasticly:Blank Group,Model Group,Drug Group and EA Group.The model rats,here made a spinal cord contusion by using a modified Allen's method.(1) Comprehensive evaluation of neural function of SCI rats by CBS.(2)Detect the SOD.MDA and Glu content in injury local tissues after injury by MTB method.(3)The injured tissues of EA Group,Blank Group and Model Group were removed.Total RNA was extracted.mRNA was purified and marked by Cy3 or Cy5 fluorescence labeling to synthesizec DNA hybridized probe that was hybridized with 27K Rat Genome Array chip.Then the chip was scanned with Lux Scan 10KA scanning apparatus.The data were analyzed by Gene Pix Pro 4.0 software system.Analysis the function of the differentially expressed genes by bioinformatics technique.(4) Detected the gene expression condition of the Capn8,Sphk1 and Cc12 by qRT-PCR.Results:Six hours after the SCI,(1)Compared with the model group,the score of CBS level in EA Group and Drug group were significant differently(P<0.05).(2)Compared with the model group,the SOD content in injury local tissues were increased in EA Group and Drug Group(P<0.05),the MDA and Glu content in injury local tissues were decreased in EA Group and Drug Group(P<13.05).(3)There were 266 differentially expressed genes in Model Group,involved in multifarious functions,such as cytotoxicity,neural tissue degeneration and necrosis,inflammation after injury,the loss of the rehabilitation and regeneration ability,and so on.There were 181 differentially expressed genes in EA Group,51 genes were not differentially expressed in Model Group,36 up,15 down.Conceming functions,such as cell metabolism,ionic channel homeostasis,immunity and defense, cytoskeleton protein,extra cell ularmatrix,cell development,etc.(4)the Capn8,Sphk1 and Cc12 gene expression was up-regulate in Model Group and EA Group.This result is in good agreement with the gene chip experiment.Conclusion:EA had protective effect on the SCI through increasing SOD content and depressing the MDA and Glu content.There were lots of differentially expressed genes in SCI rats.The rise of the Capn8,Sphk1 and Cc12 gene expression level may be correlated with the ascension of SOD and the drop of MDA and Glu.EA can regulate the changes of gene expression induced by SCI..
