Objective:The Granule of Reinforcing Vitality and Resisting AIDS (GRVARA), made ofAstragalus menmeranaceus, Codonopsis pilosula, Glycyrrhiza uralensis and so onthrough special processes, has the effect of replenishing energy, reinforcing vitality,removing blood stasis and clearing out internal toxin. Clinically it is used in thetreatment of resting phase and acting phase of AIDS characterized by "toxin, stasis inthe body and deficiency of vital energy". By means of pharmacological experiment,test GRVRA's pharmacological function of immune regulating; evaluate its efficacy;provide pharmacological basis for clinical test. Observe the combination use ofGRVARA and DDI on mice; study whether GRVRA could relieve or protect from thedamage on liver and kidney incurred by DDI. Test the maximum tolerable amount ofGRVRA extract orally taken; explore toxic reaction and its graveness; provide basisfor the dosage calculation for clinical application.Method:Pharmacological and acute toxicological effect of GRVRA is studied in this article.Mice are modeled with hydrocortisone injection; these immunosuppressive modelsare orally infused with heavy, medium or light dosage of GRVRA respectively forfour weeks; they are compared with lentinan group. Peripheral blood extracted frommice' eyeballs is tested; changing status of CD4~+, CD8~+, CD4~+/CD8~+ of T-lymphcells are observed. Their immune organ, spleen and thymus, are picked out for thecalculation of weight index. In the meantime, several models of immunosuppressivemice incurred by hydrocortisone injection are set up, in addition to DDI, they areinfused with heavy, medium and light dosage of GRVRA for a comparison with pureDDI group. By means of electron microscopy, super micro structure of liver's and kidney's cross section is observed. Mice are infused with GRVRA, whose amount isequivalent to 139 times of an average adult dosage. Acute toxicological experimentis performed to observe their weight and status after zero, seven, fourteen days.Results:1.The test of GRVRA's influence over CD4~+, CD8~+ shows that, the percentage ofCD4~+, CD8~+ and the ratio of CD4~+ to CD8~+ increased significantly in comparison withthe model group, while there is not significant difference between normal group andlentinan one.2. The test of GRVRA's influence over spleen and thymus index shows that, thetwo indices of light dosage group increase, and the immune organ restores well. Lightdosage group and normal one form a sharp contrast.3. Preliminary test of GRVRA's relieving effect against DDI shows that lightdosage of GRVRA helps to protect liver and kidney from morphological change to acertain extent.4. In the acute toxicological experiment, after infused with above mentionedamount of GRVRA, mice are found lesser activity, erected hair, dull look 20 to 30minutes later, and they restored gradually. The fourteen days has seen no death. Thereis no significant difference in weight between this group and normal one.Conclusion:After the application of GRVRA, immunosuppressive mice get improved inimmune function; percentage of CD4~+, CD8~+ and the ratio of CD4~+ to CD8~+ increases;the index of immune organ increases, which proves that GRVRA gets well inenhancing or regulating of immunosuppressive mice' immune function and isapplicable to corresponding indications. In comparison with DDI group, the liver andkidney of light dosage group are more clear in structure and all types of cells arenormal by and large; while the DDI group's are not clear in boundary, cells are in chaos, mitochondria swells and leaks. Fourteen days of acute toxicological experimentsee no toxic response, there's no significant difference in weight, which shows thatGRVRA has no significant acute toxic and adverse influence, while longer term oftoxicological experiment is needed.
