| Objective: To discuss the effects and mechanism of Winxin Granules on chronic congestive heart failure (CHF) rats. In experimental study, the model rats were built by injecting adriamycin(ADR). The experiments were carried out about the effects on the indexes of hemodynamics, serum endothelin (ET) and angiotensin II (AngII) , myocardium cell structure of CHF rats.Methods: 66 Wistar rats were tested as subjects and decided into 6 groups randomly. There was 11 rats in every groups. They are normal group, model group, treating group(Winxin Granules with great dosage, medium dosage, small dosage) and contrasting group(digoxin group). The rat model of CHF was created by injection ADR in abdomen for 6 weeks, once a week. Normal group and model group were given brine, treating group were given Winxin Granules, the contrasting group was given digoxin. After 4 weeks, all rats'heart function were mensurated by mechanocardiography. Then the concentrations of ET and Ang II in blood of rats were determined by radioimmunoassay, and myocardial cell structures in different control groups were observed under electron microscope to assess the effect of Wenxin Granules.Results: The concentrations of ET and Ang II of CHF rats were significantly increased. Wenxin Granules groups can improve the myocardial cell structures of the CHF rats to varying degrees. Wenxin Granules can increase the indexes about +dp/dt max,LVSP , and decrease the indexes about LVEDP,—dp/dt max;Conclusion: It shows that the Wenxin Granules can decrease the concentrations of ET and Ang II, and can improve the myocardial cell structures and the indexes of hemodynamics of the CHF model rats. That proves that Wenxin Granules is an effective drug in treating CHF. |
