Objective: To study the early behavior therapy to reduce cerebral palsy caused by brain damage rat.Method: 1,Establishment of model: 27 Wistar pregnant rats were randomly divided into two groups: LPS (n=21) and saline (n=6). Pregnant rats were consecutively injected with LPS (450μg/kg) or saline on gestation d 18 and 19. The pups whose gestation age was less than 22 days were preterm pups and were removed in both groups. We selected randomly saline neonatal rat group B(n=30) and LPS neonatal rat group A1(n=20),A2(n=30). 2,Early interfere: The 2 days rats( A1 )were early behavior therapy by enriched environment etc and the rats (A2,B)were fed by the routine way. 3,Neurobehavioral observation and appraising cerebral palsy: The Neurobehavior of rat was observed in A1,A2 and B groups. The items of neurobehavioral observation included sensorimotor movement (hanging test and inclined plane test), emotional behavior (open field test and resistant to capture test), posture, voluntary movement, non-autokinetic movement, muscle tension and deflected plate test. The data was analysed by the science of statistics. The cerebral palsy rats must have abnormal neurobehavior in aspects of sensorimotor movement and posture and muscle tension at the same time, accompanying or non-accompanying with the abnormity of non-autokinetic movement and emotional behavior, but the result of deflected plate test must be positive. 4,Immunohistochemistry detection: The MBP and S-100 of A2 (n=10),B(n=10) rats were detected in brain sections of 1 day rat; The MBP and S-100 of A1,A2,B rats were detected in brain sections of 25 days rat;Results: 1. 4 pregnant rats were dead after injected LPS. There were 3 pregnant rats of pre-parturition in LPS group. The rest of pregnant rats delivered 83 live mature rats and 24 dead rats in LPS group. In reverse, there were no dead or pre-parturition's pregnant rats in saline control group. The pregnant rats delivered all 46 live mature rats and no dead rats in saline control group. There were a large amount of neutrophilic granulocyte in the uterus and placenta in LPS-treated group. 2. There was obvious decrease about MBP expression in internal capsule, hippocampal, and callus in A2 group of 1 day rats, contrasted with the one in B group of 1 day rats (P<0.01); There was increase about MBP expression in A1 group of 25 day rats, contrasted with A2 group of 25 day rats (P<0.05), it was obvious increase in B, contrasted with A1 and A2(P<0.01); Contrasted with B ,there was significant increase in A2 of 1 day rats with regard to S-100 expression (P<0.01); The result of S-100 expression between A2 and B of 25 days rats is the same with the one of 1 day rats, but the S-100 expression of A1 is the middle of them, there was significant difference(P<0.01)3. The result of neurobehavioral observation indicated that there were 4 cerebral palsy rats in A1 group, 8 rats is cerebral palsy in A2 group and there was no cerebral palsy rat in B group.Conclusion: 1. Intrauterine infection can lead to brain injury of neonatal rat. 2. MBP and S-100 can become the sign of early diagnosis and the therapy effect in brain injury. 3. Early interfere could reduce cerebral palsy that was induce by intrauterine infection...
