Radix Burpleuri is one of the most common Chinese medicines. With large amount and full geographical coverage, many species of Bupleurm are locally used, so that the species still has not been clearly defined. Most radix Burpleuri was wildly collected, while the resources of radix Burpleuri is seriously destroyed, there are increasingly more cultivated ones. In order to proper application, people should systematically research the quality of cultivated radix Burpleuri. There is quality analysis about radix Burpleuri focusing on extracts in Chinese Pharmacopoeia 2005 Edition, while of saikosaponin,volatile oil should be considered as other analysis indicators for complete and reasonable analysis of Radix Burpleuri quality.Focusing on the above topics, the writer identifies the species and researches on quality of Radix Burpleuri in the North China marketsd and establishes the quality standard providing the scientific basis for Radix Burpleuri application. The methods and results are as following:(1)The investigation of the resources of products in market shows that the main resource is radix Burpleuri Chinensis, seldom is radix Burpleuri Scorzonerifolii; the majority of"nanchaihulei"are radix Burpleuri Bicauli and radix Burpleuri Angustissimi.(2)After identification of Radix Bupleurum samples by the methods of the combination of character identification and microscopic identification, the results show:①The cultivated radix Burpleuri Chinensis is deferent from the wild radix Burpleuri Chinensis in character identification and microscopic identification, they are easy to be identified, their difference should be added into Chinese Pharmacopeia;②Radix Burpleuri Scorzonerifolii,radix Burpleuri Bicauli and radix Burpleuri Angustissimi are not much different from each other in character identification and microscopic identification, usually mixed up as"nanchaihu"for sale.(3)Determine the extracts content basing on the method in Pharmacopeia:①the extracts content of wild Radix Bupleurum Chinense is 16.5±3.24%, with the ceiling of 22.6%, the passing rate is 97.6%;②the extracts content of cultivated radix Bupleurum Chinense is 15.6±3.51%, with the ceiling of 21.7%, the passing rate is 89.3%;③the extracts content of radix Burpleuri Scorzonerifolii is 25.9±4.79%, with the ceiling of 30.2%, the passing rate is 100%;④the extracts content of radix Burpleuri Bicauli is 24.39±3.79%, with the ceiling of 31.6%, the passing rate is 100%;⑤the extracts content of radix Burpleuri Angustissimi is 25.5±3.26%, with the ceiling of 28.6%, the passing rate is 100%;⑥the extracts content of Radix Burpleuri slices is 13.6±4.11%, with the ceiling of 20.3%, the passing rate is 70.0%.The cultivated radix Burpleuri Chinensis has no distinct difference from the wild radix Burpleuri Chinensis in the aspect of extracts content;radix Burpleuri Scorzonerifolii,radix Burpleuri Bicauli and radix Burpleuri Angustissimi have no distinct difference from each other; the extracts content of radix Burpleuri Scorzonerifolii is over 10% higher than radix Burpleuri Chinensis. (4)Determine the saikosaponin content by HPLC,the results are as following①saikosaponin c,a,d content of wild radix Bupleurum Chinense are 0.50%,1.90%,1.73%;②saikosaponin c,a,d content of cultivated radix Bupleurum Chinense are 0.55%,2.35%,2.27%;③saikosaponin c,d content of radix Burpleuri Scorzonerifolii are 0.15%,0.29%;④saikosaponin c,d content of radix Burpleuri Bicauli are 0.15%,0.26%;⑤saikosaponin c,d content of radix Burpleuri Angustissimi are 0.48%,1.13%;⑥saikosaponin c,a,d content of radix Burpleuri slices are 0.41%,1.17%,1.30%;⑦Saikosaponin of radix Burpleuri Chinensis could be concluded as follow,saikosaponin c% : saikosaponin a% : saikosaponin d%=6:23:21.The saikosaponin content of cultivated radix Burpleuri Chinensis is higher than that of wild radix Burpleuri Chinensis; the saikosaponin content of radix Burpleuri Scorzonerifolii and radix Burpleuri Bicauli are camparably low;the saikosaponin content of radix Burpleuri Angustissimi is comparably high.The extracts content is not related to the saikosaponin content, so that the saikosaponin content should be added as one indicator of quality control.(5)Establish HPLC fingerprints of wild radix Burpleuri Chinensis,cultivated radix Burpleuri Chinensis,radix Burpleuri Scorzonerifolii,radix Burpleuri Bicauli and radix Burpleuri Angustissimi ,confirm three main elements. HPLC fingerprints of those are different, able to be species identified and the saikosaponin c,a,d contents able to be measured in the above HPLC fingerprints condition.(6)Compare wild Radix Bupleurum Chinense with cultivated Radix Bupleurum Chinense basing on the above results, they do not differ too much in content of extracts and saikosaponin; while they differ in quantity and element of volatile oil, so that the application of cultivated Radix Burpleuri Chinense should base on standard cultivation and quality.The main resources of radix Bupleurum are radix Burpleuri Chinensis, the reserches show the distinct difference of the cultivated radix Burpleuri Chinensis and the wild radix Burpleuri Chinensis in character and microscopic identification and the minor difference in extracts content and saikosaponin content, HPLC fingerprints; while the content and components of volatile oil are different. The cultivated radix Burpleuri Chinensis is valuable in application and economy. It is highly recommended that the identification and the quality control of the cultivated radix Burpleuri Chinensis should be added into Chinese Pharmacopeia, and also the microscopic identification, saikosaponin content measuration and HPLC fingerprints. The extracts content of radix Burpleuri Chinensis is 15.6%, saikosaponin c,a,d contents are 0.50%, 1.90% and 1.73%.The extracts content and saikosaponin content of radix Burpleuri Chinensis planted in Shannxi,Shanxi,Hebei,Gansu are not distinctly different, which show the planted location has no much influence with the quality of Radix Bupleurum. Radix Burpleuri Scorzonerifolii is seldom in market, which is distinctly different from radix Burpleuri Chinensis in character, microscopic identification, extracts contents and saikosaponin content, HPLC figerprints and the content and components of volatile oil, it is recommended that radix Burpleuri Chinensis and radix Burpleuri Scorzonerifolii should be saparatedly quality controlled. The extracts content of radix Burpleuri Scorzonerifolii is 25.9%, saikosaponin c,d contents are 0.15% and 0.29%.The radix Burpleuri slices are mainly radix Burpleuri Chinensis, the extracts content of which is 13.6%, saikosaponin c,a,d contents are 0.41%, 1.17% and 1.30%。Radix Burpleuri Bicauli and radix Burpleuri Angustissimi are sold as radix Bupleuri Scorzonerifolii in market with high market share. The extracts contents, saikosaponin content, fingerprints, the volatile oil contents and volatile oil components of radix Burpleuri Bicauli and radix Burpleuri Angustissimi are similar with that of radix Bupleuri Scorzonerifolii, able to be used as new resources.The writer adjusts and supplements the quality standards of that in Chinese Pharmacopeia, and establishes the draft of quality standard of radix Burpleuri Bicauli and radix Burpleuri Angustissimi.
