Syndrome is the summary of the development changing during the different periods of the disease and the core of treatment based on syndrome differentiation. Syndrome research is the focus of Chinese Medicine research. Syndrome is indivisible with the disease, and they are dependent on each other, which is the acknowledged connection between disease and syndrome in Chinese Medicine.Acute cerebral infarction is a common critical and dangerous disease, Qi-deficiency and Blood-stasis is a familiar syndrome which may be expressed through the whole course of the disease. This study focused on Qi-deficiency and Blood-stasis syndrome of acute cerebral infarction. We established a model combined cerebral infarction with syndrome of Qi-deficiency and Blood-stasis using intraluminal thread occlusion after exhausting swimming. Qi-deficiency was taken as the initial factor or key factor running through the whole procedure of the model establishment. We observed the correlative macro-signs and the biological changes dynamically, searched the relationships between macro-signs and biological indexes, and set up the evaluation system on this model.Objectives: To establish a model cerebral infarction with syndrome of Qi-deficiency and blood stasis using intraluminal thread occlusion after exhausting swimming, discuss the method of the model establishment of disease conbining syndrome. To study the correlation between macro-signs and the biological indexes through the observing of the common state, neurological deficit scores and biochemical criterions .To set up the evaluation system on the model.Methods: 64 male SD rats weighing approximate 200g were randomly divided into fore groups: normal group, operation only group, swim only group, disease combining syndrome group, 16 rats every group. Normal group and operation only group were feed as normal, without interference. Swim only group and disease combining syndrome group were practiced exhausting swimming every day for 3 weeks. Then cerebral ischemia 2 hours and reperfusion 24 hours modal were established using intraluminal thread occlusion in operation only group and disease combining syndrome group. The weight, appetite, activity and death rate after operations were compare between the swimming rats and no swimming rats. The common state before and after the operation were observed. Neurological deficit scores were assessed according to Zea-longa method 6 hours after the operation. Morphologic changes was observed, and biochemical criterions including the activity of superoxide dismutase (SOD) and nitric oxide synthase (NOS), the content of malondialdehyde (MDA) and nitric oxide (NO) in brain were examined, 24 hours after operation.Results: Compared with the normal group and operation only group, the weight, appetite, activity of swim only group and disease combining syndrome group were much lower, but the death rate and the neurological deficit were higher. The morphologic change of brain tissue in operation only groups was milder than in disease combining syndrome group. Compare with the normal group, the activity SOD in brain tissue were all decreased in swim only group, operation only group and disease combining syndrome group, and the decrease was much significant in the later two groups (P<0.05). The content of MDA in brain was significantly increased in disease combining syndrome group than other groups (P<0.05).Conclusions: The establishment of syndrome model is a pressing problem in the field of Chinese medical research. The method which we used to establish a model cerebral infarction with syndrome of qi-deficiency and blood stasis may develop our thinking train and method of the model establishment of syndrome manifestations of Chinese medicine. The results show that the macro-signs and biological indexes of this model are according with the characters of cerebral infarction with syndrome of qi-deficiency and blood stasis. So combine with other disease and syndrome models, we can study the essential and the material function about Syndrome.
