Acupuncture Pretreatment On Formalin Pain In The Rat Central Nervous System Of C-fos, Hsp-70, Il-1 Impact

Intention: For doing further study on the theory of ache acupuncture therapy, the models ofFormalin-ache-rat, which were did acusectors "foot three miles" and "loop-jump"pretreatment, were built in this paper. For each rat in different period, we observed the achereaction, the expression of c-fos gene and hsp-70 by RT-PCP mensuration, the change of IL-1content of rat brain histiocyte by immunity histochemical mensuration. We discussed inherenttheory of acupuncture regulating effect from points of view of different spinal cord and brainlevel and animal behavior.Method: (1) "After (foot) three miles" and "loop-jump" pretreatment by acusectors. (2)Build the ache-rat models by injecting Formalin liquor to fight after sole of rats. (3) Build thecontrast models by injecting physiological brine. (4) Record the times of licking and biting itsfight after sole of each rat to mensurate the ache reaction. (5) Observe the expression of c-fosgene and hsp-70 by RT-PCP mensuration. (6) Mensurate the IL-1 content of rat brainhistiocyte by immunity histochemical coloration method.Results: (1) In 0~10 minutes and 20~40 minutes, both groups of rats being injectedFormalin liquor and physiological brine lick their claws. By the time extend, the times of thefirst group of rats licking their claws are increased, while the times of the other groupdecrease. After acusector pretreatment, the rats being injected Formalin liquor lick claws lessthan the rats without pretreatment. (2) The group of normal rats have no c-los gene expression,and the contrast acusector pretreatment group have some c-los gene expression. The achegroup of rats being injected Formalin liquor have lots of c-fos gene expression, and thecontrast acusector pretreatment ache group have more c-fos gene expression. (3) The group ofnormal rats have no hsp-70 gene expression, and the contrast acusector pretreatment grouphave some hsp-70 gene expression. The ache group of rats being injected Formalin liquorhave lots of hsp-70 gene expression, and the contrast acusector pretreatment ache grouphave more hsp-70 gene expression. (4) IL-1 electropositive nerve cells serosity of the group ofnormal rats and its contrast group of acusector pretreatment rats are light colorated and few.The electropositive nerve cells of the group of rats being injected Formalin liquor are darkcolorated and numerous. While the IL-1 electropositive nerve cells of the acusectorpretreatment group of rats being injected Formalin liquor are a little dark colorated andnumerous, the electropositive degree of the that group is depressed compared to this contrastgroup without pretreatment.Conclusions: Acusector pretreatment could activate the horizontal anti-ache system of boththe spinal cord and upside of acanthi, could increase the ache threshold and decrease thelicking reaction of Formalin stimulus. Acusector pretreatment could intensify the gene expression of c-fos and hsp-70 in pain, and accordingly enhance the anti-ache matterexcretion and protecting function. Acusector pretreatment could decrease the IL-1 expressionleading by ache stimulus and inflammation. So it does some work on abirritation of centralnerve system. In conclusion, the action of acusector on abirritating pains is realized thoughregulating anti-ache matters of spinal cord and upside of acanthi. And the acusector alsoregulates the expression of gene and albumen, regulates the reaction of behavior ache, andeases pain efficaciously.
