Ningxian Decoction Epileptic Kindled Rat Model Of Eeg And Gene Expression

Objectives: To further explore an improved method of kindling with rapidly recurring hippocampal seizures; To further explore the effects of Ningxian decoction on convulsion in epilepsy and the expression of c-fos protein in hippocampal seizures of the mice with experimental epilepsy induced by the method.Methods: Chronic seizure of epilepsy was induced by an improved method of kindling with rapidly recurring hippocampal seizures with the weight of 230-300g, and the mice at 4, 5 degree were selected as the objective of study. The mice with experimental epilepsy were randomized into 5 groups: model group, western group, Ningxian high dosage group, Ningxian middle dosage group and Ningxian Low dosage group, another 6 normal mice were selected as the control, following experiments were proceeded respectively: Firstly, the study of behavior was performed. Secondly, 28 days after therapy, evoke potential epilepsy, at the same time record EEC Thirdly, ABC immunohistochemical decoration method was adopted to study the expression of c-fos protein in the hippo region. Finally, quantitative image analysis was carried out with medical image analysis system. Results: 1. The behavior study showed that Ningxian decoction and VPA could effectively inhibit the seizures induced by kindling with rapidly recurring hippocampal seizures (p<0.05). Among western group, Ningxianhigh dosage group and Ningxian middle dosage group, the three treatment groups had no significant difference in the number of the mice with seizures, degree of seizures and latent period (p>0. 05). Between Ningxian low dosage group and the three groups were significant different (p<0. 05). 2. EEG displayed that the four treatment groups could improve the ECG of experimental epilepsy. Among the four group, there was no significant difference (p>0.05).
