| Objective To contrast the curative effect of Rosiglitazone, Metformin and Rosiglitazone plus Metformin on insulin resistance in patients with type 2 diabetes mellitus.Methods Using a randomized 3 group design, ninety type 2 diabetes mellitus patients that was inadequately controlled with diet and oral sulfonylureas. were selected and were randomized to receive Metformin, Rosiglitazone, or Rosiglitazone plus Metformin. for 12 weeks. Fasting plasma glucose (FPG), 2 hours postprandial plasma glucose (2HPG), hemoglobin A1C (HbA1C), body mass index (BMI), systolic blood pressure (SBP), diastolic blood pressure (DBP), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL), fasting plasma insulin (FIns) , body height(cm), body weight(kg) were recorded before and after therapy. The insulin resistance (Homa-IR) andβ- cell function (Homa-β) of the patients were evaluated using homeostasis model assessment (Homa).Results Compared with pre - treatment values, FPG,2HPG,HbA1C,SBP,DBP,TC,Homa-IR decreased evidently , and Homa-βincreased significantly in three groups. However, LDL,BMI decreased obviously in the metformin group, TG,FIns decreased obviously in the rosiglitazone group, LDL,BMI,TG,FIns decreased significantly and HDL increased significantly in the metformin plus rosiglitazone group. After 12 week of treatment: FPG,2HPG,HbA1C had statistically significant differences between rosiglitazone plus metformin group and metformin group, meanwhile FIns,Homa-IR in rosiglitazone plus metformin group had statistically significant differences compared with the rosiglitazone group and the metformin group. There were statistically significant differences between rosiglitazone and metformin in FIns,Homa-IR. There were no statistically significant differences among the three groups in SBP,DBP,TC,TG,LDL,HDL,BMI,Homa-β.Conclusion Both rosiglitazone and metformin can decrease insulin resistance, but rosiglitazone is bettet than metformin,.and rosiglitazone plus metformin have synergistic action. |
