Background: Chronic prostatitis (CP) is a common disease in male genitourinary system. To this day, its cause remains to be unclear. At present, there has not been received method about establishing experimental autoimmune prostatitis animal model (EAPAM). The research about EAPAM is still less both at home and at abroad. Therefore, it is necessary to copy valid and stable animal model of CP to explore the pathogenesy of CP.There is short of effective medicine in clinical study now. Thus, it is of great importance to search for the mechanism of CP and develop available Chinese patent medicine. Based on main symptoms local pain such as hypogastric zone, the waist and perineal position and prostate hardening , tenderness by digital rectum examination(DRE), it is in line with syndrome of blood stasis of TCM. Combinedwith pathological change feature of prostate ------obstruction of glandular cavity,stagnation of prostatic fluid and interstitial fibrosis, the principal of promoting blood circulation and removing toxic substances was set up integrated macroscopic syndrome differentiation and microcosmic syndrome differentiation. Thus, Dan Pu capsule (DP) are prepared by Prof. Liu You Fang, famous urinary surgery specialist integrated traditional and western medicine.My tutor, Prof. Zhang Ya Qiang, is the academic heir of Prof. Liu You Fang. The result suggests thatthe principal of promoting blood circulation and removing toxic substances is very important to treat CP;DP capsule has notable curative effect, which can improve clinical symptom obviously.Purpose: The study aims to establish EAPAM to explain the immunologic mechanism of DP capsule treating CP, which may supply theoretical and experimental foundation of the principal of promoting blood circulation and removing toxic substances in the therapy of CP.Materials and methods: Selecting Wistar rats, SD rats and C57BL/6 mice as experimental animal, prostatic tissue was taken out under germfree conditions. After being weighing and cleaning, it was added into 0.5% TritonX-100 normal saline. Then the homogenate was made by glass homogenizer. Centrifuged with high-speed centrifuge, the supernatant fluid was obtained, and then the protein level was measured. Thus, EAPAM was duplicated by injecting subcutaneously complete freund adjuvant (CFA) with multi-points. Then, the effect of DP capsule on CP was to be found. The 60 animal model of Wistar rat were divided by 6 groups: high dose group of DP capsule(HD), middle dose group of DP capsule (MD) ,low dosage group of DP capsule (LD) ,Qian Lie Tai pill group(QLT), the model group and the normal group. The rats were lavaged in HD, MD, LD and QLT and the remnant rats were administrated by 0.9% normal saline. They were sacrificed 56 days later. The gross shape was viewed by macroscopic observation. Finally, the level of IL-8, IL-10, TNF-a and PGE2 in serum and prostatic tissue were detected in different groups by means of enzyme linked immunosorbent assay.Results: The clear inflammation is found in prostatic tissue of the model group. Compared with the normal group, the level of IL-8 & TNF-a are on the rise in serum and prostatic tissue of the model group. Meanwhile, IL-10 & PGE2 alse arise in prostatic tissue of the model group. There are striking differences between the twogroups above (P<0.05 or 0.01) .Compared with the model group, the level of IL-8 & TNF-a are on the decrease in serum and prostatic tissue of the MD group. Likewise, IL-10 & PGE2 reduce in prostatic tissue of the MD group. There are striking differences between the model group and the MD group as well (PO.05) .Conclusions: It is effective method to establish EAPAM by injecting prostatein refining liquid and CFA. Autoimmune factors take part in the process of CP and cell mediated immunity may pay an important role in the mechanism. The principal of promoting blood circulation and removing toxic substances is vital in the treatment of CP. DP capsule is available Chinese patent medicine, which can improve local inflammation of prostate and regulate local immunity state.
