Prostatitis in one of the most commonly encountered diseases at Urology clinic, which accounts for 8%-25% of the total outpatients. With the characteristics of various symptoms, long-course therapy, easy recurrence, most patients are tortured both mentally and physically. Now there are lots of researches on prostatitis, however the mechanism of the chronic prostatitis is still not clear. More and more evidences show that Th cells and T reg cell play an important role in prostatitis, which leads many scholars to pay more attention to the immunological mechanism of chronic prostatitis.DanPu Capsule is invented by the Urology Department, Guang'AnMen hospital based on the year-on-year clinical experiences on prostatits treatment. It is already proved that DanPu capsule could surpress the expression of IL-Iβ, IL-2, IL-8, IL-10and this study tries to investigate DanPu Capsule's regulatory effect on Th cell and T reg cell in EAP model rats.This essay consists of two parts:literature review and experiment research.Literature review:To summarize the recent researches on prostatitis both in western medicine field and TCM field.Experiment research:Objective:To investigate DanPu Capsule's therapeutic mechanism on prostatitis by observing and analyzing DanPu Capsule's influences to the Th cell and T reg cell in EAP model rats. Method:42 male wistar rats are divided randomly into 4 groups:normal group, EAP model group, DanPu Capsule group and QianLieTai group.10 rats for each group. Purified rats prostatitis protein solution mixed with FCA was injected subcutaneously to all the rats except normal group at the start, and prostatitis protein solution mixed with FIA was injected on 21st day. Normal group and EAP model group were given physiological saline intragastrically and DanPu Capsule group and QianLieTai group were intragastrically given DanPu Capsule solution QianLieTai pill solution respectively. All the rats were sacrificed after 8 weeks with blood and prostate tissue specimens analyzed. Result:compared to the normal group, the prostate tissues of EAP model rats are found infiltrated by the inflammatory cells observed through light microscope and the gland cells are disorganized observed through electron microscope. The CD4+CD25+Treg decreased at the meanwhile IL-4 also decreased and IFN-y increased. But in DanPu Capsule group, only a few inflammatory cells can be observed and most cells remain normal structure. Compared to the EAP odel rats, IFN-γdropped dramatically and Treg cells and IL-4 increased notably, which shows the balance of Th1/Th2 approached to the normal level. Conclusion:DanPu Capsule could give a regulatory effect on the balance of Th1/Th2 of the EAP model rats, and lead balance to a normal level so as to effectively release the inflammatory reaction of the local tissue.
