Shenfu Injection Of Rats With Experimental Heart Failure Treatment Effects And Mechanisms

Objective : By observing the effect of shenfu injection to the hemodynamics, myocardial apoptosis and the plasma TNF-α,IL-6 and sFas of heart failure rat, to explore the function mechanism of shenfu injection in the treatment of congestive heart failure.Methods: Rat model of heart failure after myocardial infarction was established by ligating the left anterior descending branch of coronary artery. After operation for one week, rats were randomized into three groups:Sharm-operated group (the surgical process was placed but not ligated,n=14),model group of heart failure(CHF,n=13) and shenfu inje- ction group(n=11).The rats of shenfu injection group were treated with shenfu injection(6.2ml/kg/d) and the other two groups were treated with equal 0.9% sodium chloride for 4 weeks.Four weeks after the initiation of therapy,hemodynamic studies were performed in each group.The right common carotid artery cannulatio entered into left-ventricle,then BL-420 biological experiment system recorded all of LVSP, LVEDP, +dp/ dtmax and -dp/dtmax from rats.These rats were killed after measuring the hemodynamic parameters.The hearts were harvested and made to paraffin sections.Apoptotic myocardial cells were detected by TUNEL Method.The expressions of Fas, FasL, Bcl-2 and Bax were analyzed immnohistochemically with their antibodies.Finally, we used ELISA to detecet the circulation level of TNF-α,IL-6 and sFas. Results:1. Hemodynamics detecting result: compared to sharm-operated group, the numeric value of LVSP, +dp/dtmax and -dp/dtmax of rats in the Heart failure group notably declined(P<0.05or P<0.01), and LVEDP notably rose(P<0.01).In shenfu therapeutic group,the above hemodynamic parameters significantly improved(P<0.05).2.Left ventricular weight index: compared to sharm-operated group, the left ventricular weight index of rats in the Heart failure group notably rose(P<0.01),but LVWI improved in shenfu therapeutic group(P<0.05).3.The apoptosis in myocardial cell significantly increased in heart failure group and shenfu injection group.But the apoptosis cells in...
