Objective In order to set up a post-stroke rat model and investigate the influence of "supplement Qi and activating Blood circulation " formula on level of IL-6 mRNA after acute cerebral ischemia.Method Set up a focal cerebral ischemia rat model by occluded right middle cerebral artery of rat with Electrocoagulation method (MCAO). Give drugs to rats before making model ,once a day. MCAO rats are devided into 4 groups randomly as following :High dose group, middle dose group, low dose group, cyclophosphamide group . Imitate operation group , model group and normal group as contrast groups. After making model of MCAO ,all rats are observed with Longa 5 grades of nerve function .Intervention period are 24 hours , 48 hours , 72 hours respectively and detect IL-6 mRNA expression level with RT-PCR.Results The model group IL-6 mRNA expression level increase and after 24 hours and decrease after 48 hours and 72 hours . In "supplement Qi and activating Blood circulation " groups, with increasing of dose , the expression level of IL-6 mRNA is decreasing .The effect of high dose group is close to the effect of cyclophosphamide group .The normal group and imitate operation group keep low IL-6 mRNA expression level.Conclusion "supplement Qi and activating Blood circulation " formula andcyclophosphamide can lessen nervous function and reduce volume of cerebralischemia .Normally ,IL-6mRNA expression is low. After acute cerebral ischemia"supplement Qi and activating Blood circulation " formula can inhibit MCAO model ratinflammation response and depress the level of IL-6 mRNA . The effect of highdose group is close to the effect of cyclophosphamide group. So we can speculate that"supplement Qi and activating Blood circulation " formula can control inflammatorycell gene to improve nervous function and ischemia volume. |
