Objectives: To investigate the changes of gingival crevicular fluid (GCF) volumn and activity of Aspartate aminotransferase (AST) and Alkaline phosphatase (ALP) in gingival crevicular fluid during porcelain fused to metal (PFM) crowns restoration. Relation between the activity of AST, ALP and the health of periodontal tissue was discussed, and the effect of location of crown margin on periodontal tissue was explored.Methods: 19 patients with PFM crowns were selected in this study, the Randomized Controlled Trials (RCT) method was used in this study and the subjects were divided into two groups randomly: subgingival group and gingival crest group. Under circumstances of strictly limiting crown axial contour, margin discrepancy and periodontal conditions, porcelain-fused-to-metal crowns , A total of 30 teeth were screened, subgingival group was consisted of 16 teeth, gingival crest group was consisted of 14 teeth. GCF was collected from mesial sites in two groups before the tooth preparation. The volumn of GCF and the activity of AST and ALP were regarded as the baseline. At the point of 2 weeks. 1 month, 2 months and 3 months after cementation , GCF was collected for longitudinal investigation and comparison between subgingival group and gingival crest group. The data of GCF volumn and the activity of AST and ALP was analyzed statistically by SPSS 10.0.Results:1. In gingival crest group , after inserting crowns , the volumn of GCF , GCF-AST activity and GCF-ALP activity was no significantly change on 2 weeks, 1 month and 2 months , but the volumn of GCF and GCF-AST activity was significantly increased on 3 months.2. In subgingival group, after inserting crowns , the volumn of GCF and GCF-AST activity was significantly increased on 2 weeks, 1 month , 2 months and 3 months.
