Schizophrenia is one of severe psychosis occurring frequently, whose cause is unknown, and mainly appears psychomotility detaching from reality and loss of unity and integrality of whole spirit. Its symptoms vary significantly, so that it heavily impairs social adaptability of the patients. Through recently 50 years, The treatment of schizophrenia has gained enormous improvement, but antipsychotics-caused severe side-effects influences quality of life and drug compliance of patients. The traditional Chinese medicine has believed that schizophrenia could be classified with "Dian Syndrome(depressive state)" and "Kuang Syndrome(mad state)" based on negative and positive symptoms of schizophrenia. The Chinese medicine has advantage on the facts that combination with APDs may significantly alleviate various adverse reactions of APDs and remarkably improve negative symptoms and cognitive disorder of schizophrenia patients. We had clinically applied DaWei capsule to treat schizophrenia and observed satisfactory effects. The general pharmacological action of DaWei capsule to central nervous system and behavioral pharmacological action of schizophrenia animal models was studied, and neurotransmitters (including 5-HT, NE, DA) content in related brain region and serum in schizophrenia animal models with negative symptoms were measured by fluorospectrophotometry, and lipid peroxide (LPO) level in serum was measured, and hemorheology changes was observed. 1 The general pharmacological action of DaWei capsule to central nervous system 1.1 The influence on spontaneous activity of mice The spontaneous activity of mice was measured by DigBehv Spontaneous Activity Video analysis system before and after DaWei capsule administration. The results show that total distance of the mice in large-dose DaWei capsule and diazepam group is significantly decreased compared with control group after 3-day administration (P<0.01); total distance of the mice in large-dose DaWei capsule and diazepam group is significantly decreased (P<0.01), while that in middle and small-dose groups is decreased with no significant difference as self-comparison before and after administration. 1.2 The effects on mice sleep duration caused by sodium pentobarbital with DaWei capsule After 3 days administration, the mice were given with sodium pentobarbital (40mg/kg), ip, and synergetic effect was observed. The results show that mice sleep latent period in large-dose DaWei capsule (P<0.05) and diazepam (P<0.01) groups is significantly decreased, while that in middle and small-dose groups is decreased with no significant difference compared with that in control group; mice sleep duration in large and middle-dose DaWei capsule (P<0.05)and diazepam (P<0.01)groups is significantly increased, while that in small-dose DaWei capsule is increased with no significant difference. 1.3 The effects on mice convulsion caused by pentetrazole with DaWei capsule After 3 days administration, the mice were given with pentetrazole (110mg/kg), ip, and convulsive effect was observed. The results show that mice convulsion latent period and survival time in phenytoinum group is significantly increased, while that in large, middle and small-dose groups is no significant difference compared with that in control group; mice mortality in phenytoinum group is significantly decreased, while that in large, middle and small-dose DaWei capsule groups is no significant difference. 2 The effects on behavioral pharmacological action in schizophrenia animals models with DaWei capsule 2.1 The effects on spontaneous activity in amphetamine-induced schizophrenia mice models with positive symptoms After 3 days administration, the mice were given with amphetamine (6mg/kg), ip, and spontaneous activity was observed. The results show that mice total distance in clozapine (P<0.05) and haloperidol (P<0.01) group is significantly decreased, while that in DaWei capsule groups is no difference. 2.2 The effects on immobility time(IT) in forced swimming test(FST) in ketamine-induced schizophrenia mice with...
