Objective:To determine the effect of negative emotions on allergic rat model byexamination cytokines in serum and NK cells count in peripheral blood,theexpression of NK2-R in lung and observe the differentials during inhaledglucocorticoid.Methods: forty healthy male SD rats were randomly divided into four groups, groupA as the control group, group B as the allergic group, group C as thestressed-allergic group,group D as the glucocorticoid-treated,stressed-allergicgroup.Male rats were sensitized and challenged by OVA except for the rats in controlgroup to establish an allergic model,forced swimming Test was given at the sametime as OVA sensitize in group C and group D,glucocorticoid was inhaled at thesame time as OVA challenge in group D. Symptoms related to asthma and generalappearance(such as eating, drinking water)of all rats were observed during theexperiment.Airway peak pressure was measured with four-lead graph system,HEstaining was used to observe the inflammation of the lung tissues,the levels of NKcells in peripheral blood were detected by flow cytometry,the cytokines IL-4,GC,IFN-γ in the serum were determined by ELISA,NK2-R mRNA in lung tissueswas evaluated by RT-qPCR.Results:Group A showed low levels of baseline NK2-R expression in lung tissuesand difficult to obtain.Compared with group A,Pulmonary inflammation score,Airway peak pressure,the serum IL-4levels were higher in group B(P <0.05,respectively)and NK2-R mRNA in lung tissues were higher than group A,while theserum IFN-γ levels and the counts of NK cells in peripheral blood were lower thangroup A(P <0.05,respectively),yet the GC in the serum showed no statisticallysignificant difference between group A and B (P>0.05).The Pulmonaryinflammation score,the serum IL-4,GC and airway peak pressure were higher ingroup C than those in group A and B(P <0.05,respectively),while the serum IFN-γlevels and the counts of NK cells in peripheral blood were lower than in those groups (P<0.05,respectively),NK2-R mRNAin lung tissues was greater compared withgroup B(P<0.05).Compared with group C,Pulmonary inflammation score,airwaypeak pressure,the serum IL-4levels and NK2-R mRNA were reduced in group D(P<0.05,Respectively),the IFN-γ levels and the counts of NK cells in peripheralblood were increased(P<0.05,Respectively),the GC levels in the serum showed nostatistically significant difference between group D and C(P>0.05).Compared withgroup B,airway peak pressure,the serum IL-4levels were reduced in group D(P<0.05,Respectively),the IFN-γ levels were increased(P<0.05),while pulmonaryinflammation score,the serum GC levels, the counts of NK cells in peripheral bloodand NK2-R mRNA in lung tissues, no differences were found between group D andgroup B (P>0.05, respectively). Compared with group A, pulmonaryinflammation score,airway peak pressure,the serum GC levels were increased ingroup D(P <0.05,Respectively),the counts of NK cells in peripheral blood weredecreased,the serum IL-4and IFN-γ levels between group D and A were notsignificantly different(P>0.05,Respectively).Conclusion: Negative emotions are often associated with exacerbation of allergicpulmonary inflammation.Negative emotions usually cause activation of thehypothalamic-pituitary-adrenal (HPA) and a subsequent release of glucocorticoids.Negative emotions also effect the balance of Th1/Th2cytokines,increase theexpression of NK2-R in the lung tissue and reduce NK cells in peripheralblood,which maybe important in the seriousness of asthma.ICS can significantlycorrect the balance of Th1/Th2cytokines,decrease NK2-R expression in lung tissuesof asthmatic rats.Maybe increase peripheral blood NK cell numbers via any otherways.
