Chronic blood-lacked diseases of lower limbs are plentiful in theperipheral vascular. Despite the therapy in this aspect have moredevelopment, still some patients' vessels don't regenerate depend onoperation and interposition because of various factors.Thesepatients'symptom still happen repeatedly in maximum medication,even thesepatients have the possibility of cutting lower limbs.In recently years,ABMCT that is simple on operation and doesn't appear immune repulsion andEPC medication become preferable treatment to the chronic blood-lackeddiseases of lower limbs. ABMCT is a new treatment to the chronic blood-lacked diseases in recentyears. First the patients'bone-marrow stem cells that are transplantedto blood-lacked places of the limbs proliferate and become vascularendothelial cells. The vascular endothelial cells build abundant lateralsmall blood vessels that can ameliorate local blood circulation.But thepolarization and control mechanism of bone-marrow stem cells is still notclear. In clinic work we find that some factors such as superior ages,longer suffering time and more syndromes can affect the clinic curativeeffect through decreasing bone-marrow stem cells,weakening stem cells'polarization ability and destroying proliferation environment. Interrelated investigation indicate that "promoting Bloodcirculation for tissue regeneration" "supplementing Qi for activatingBlood circulation"and herbal medicine can promote vascular regeneration.Herbal medicine can increase vascular regenerative cell factor through:① increasing BM-MNC and enhancing its immunocompetence; ② inducingantibody from B cells and enhancing its immunocompetence. Herbal medicinehas excellent curative effect to sphacelus through decresing bloodviscidity,counteracting blood solidification ,dissolving fibrin,dilating blood vessel, increasing blood stream and regulating immunefunction. Arteriosclerosis and diabetes vascular syndrome can result in ischemiaof lower limbs that belong to "Bi-syndrome of the vessels","sloughingJu"in the tradintional chinese medicine. In this opinion , weekness dueto chronic disease, Blood stagnation due to Qi Deficiency.And the patientsare almost old persons with deficiency of Qi and Blood. According to ourexperiences, this disease almost belongs to syndrome of Qi Deficiency andBlood stagnation, and the application of supplementing Qi for activatingBlood circulation acceieves good effect in clinic. This investigation is base on theory of "supplementing Qi foractivating Blood circulation". we select Astragalus membranaceus andPanax notoginseng because of good clinic curative effect. In thisexperiment we research two herbal medicines' effect to promote bone-marrowstem cells to transform and increase EPC in vitro. This can provideexperimental evidence for ABMCT combined with herbal medicine curingchronic blood-lacked diseases of lower limbs. And also we will investigatethe mechanism of EPC proliferation. Method:Collect the marrow blood of patients blood-lacked in lowerlimbs, seprate BM-MNC, and increase the cells with different condition.Morphologic observation , and measure the ratio of CD34+ cells after threeweeks. Astragalus membranaceus injection and Xuesaitong powder arerespectively divided into 3 groups randomly: ①high dosage group; ②middosage group; ③lower dosage group. And also control group is set up. Result:⑴From the thmorphologic observation, the shuttled cells isabandunt, karyon be placed in the middle and collect together. With timegoes by , cells that is shuttled and polygonal become monolayer,arrangeas vortex.These indicate the cells have more morphologic feature is liketo endothelial cells. ⑵the ratio of CD34+ cells: Compared with controlgroup, Astragalus membranaceus lower and mid dosage group , Panaxnotoginseng mid and high dosage group can increase CD34+ cells percentageevidently. But Astragalus membranaceus high dosage group and Panaxnotoginseng lower dosage group haven't statistic meaning compared withcontrol group. Astragalus membranaceus mid dosage group and Panaxn...
