Angongning patch is consisted of Scutella ribaica lensis Georgi. Based on the Angongning capsule that have a good clinical effect , we extract the effective ingredient from Scutella ribaica lensis Georgi with the advanced method of abstraction and refinement of traditional Chinese medicine. We reform the capsule to the patch, and afford the date to preparation report after a series of experiments.At first, we comfirm the effective composition of Angongning patch by pharmacology experiments . the condition of extraction and refinement and Pharmaceuticals were researched by orthogonl design and the optimum preparation process were determined. In quality standards studies, the study on TLC identification method for baicalin and wogonin was carried out. Then we established the content determination method of Angongning patch as the marker of baicalin and wogonin. Determinated the content of baicalin and wogonin by HPLC, we research the release and penetration of the patch. We also studied the preliminary stability of Angongning patch at normal temperature tests .To optimize the extraction process of Huangqin herb, the optimum extraction condition were investigated by the orthogonal design experiment . In yields of extracts , baicalin and wogonin were determinted as the content indexes. The optimum extraction condition was obtained : decocting the drug 3 times, 12 folds of water extracting for 1. 5h after boiled once; extracting the remnant of drugs 3 times, adding 5 folds of 85% alcohol extracting for 1h after boiled. The optimized process is stable and highly efficient.To optimize the refinement process of Huangqin extracts, different methods were applied to different effective composition. The optimum refinement condition of Huangqin extracting by water were investigated by the orthogonal design before absorption of macro reticular resin,The optimum refinement condition obtained : concentrating the density of the liquid to 1.5, adding alcohol to 40%, depositing 24h, centrifugalizing.To study the technological parameters of the purification process of hangqin extraction with macro reticular resin. The adsorptive characteristics and elutive parameters of the process were studied by taking the elutive and purified ratio of flavonoids as markers. results: the AB-8 macro reticular resin was suitable to adsorpt flavonoids of huangqin by choosing different styles of macro reticular resin; The optimum adsorption condition were investigated by the orthogonal design, and we confirmed the quantity of adsorption and the condition of alcohol eluting by a series of experiments. The process of applying macre reticular resin to adsorb and purify huangqin is feasible.To optimize the refinement process of Huangqin extracts by alcohol, we reseached different methods of extraction .We confirmed the method of extraction by Ethylacetate was valid. Then we enlarged the experiment and make sure the project was feasible.The optimum of the making process of the patch was investigated by the orthogonal design. the result: 0. 4g Glycerol, 0.3g Monoglyceride , 0. 3g Paraffin, 0. 4g Vaseline and 0.1g Sodium lauryl sulfate were made up of a piece of the patch.The experimental results had shown that TLC spot of Scutella ribaica lensis Georgi were very clear in the perpartion and had repeatability. The content baicalin is not lower than 100mg in a piece of patch and the content wogonin is not lower than 8mg in a piece of patch. The stabilities, sensitives,recoverity conformed to the demand of qualitative analyses. These method can be adopted as a quality content method.The research of the release and penetration rate of the Angongning patch: with the time prolonging, the content of baicalin and wogonoside which was releasing and penetrating from the patch was increasing. The content of wogonoside that was released in the water within 80min amounted to 50. 1%, the rate of penetration amounted to 25. 4%.The result of preliminary stability indicated that the result of characters,indentification, test, contentdetermination unchanged in three mont...
