Leukemia is a Primary Hematologic in building-blood organize, it is a malignancy from haematology and harms badly the health of mankind. The hematopoietic stem cell autograft has become a optimal approach, and purifying in vitro is the key of successful auto-transplantation.In the first part of the dissertation, we narrated the research meaning and content. In the second part, we summarized the ALA-PDT and the cancer therapy based on nanoparticles. In the third part, we given a chief narration about the prindipium of MTT assay and FTIR spectroscopy. In the forth part, we did some experimental research on the infection of TiO2 and TiO2Nx, in the size of nanometer (TN) to the HL60 cell activity by MTT assay. In the fifth part, we analysed the change of conformation and Protein Secondary Structure of HL60 cells after the action of ALA, TN and ALA-PDT by FTIR. In the last part, the whole dissertation was summed up.The major achievements made in this dissertation were as follows:(DThe feasibility was analysed that ALA-PDT based on NT can improve the sensitivity killing HL60 cells.(2)We did some experimental research on the infection of TiO2 and TiO2Nx in the size of nanometer (TN) to the HL60 cell activity according on different conditions, such as the time cultivated, the final concentration added TN, the cell inoculated , light catalyse and so on.(3)Changes of conformation and Protein Secondary Structureof HL60 cells were analysed after the action of ALA, TN and ALA-PDT by FTIR.(4)The relation between the cell activity and conformation was studied on the level in molecule structure, and a new optical way inspecting the cell activity is put forward.
