The Huayu Qutan Granule On Experimental Animal Atherosclerotic Vascular Endothelial Function, Platelet Activation

Objectiv To investigate the intervention of Resolving stasis eliminating phlegm in the development and progress of atherosclerosis(AS).Methods 32 healthy male New Zealand rabbits were divided randomly into 4 groups:(1) normal group(n=8)were fed with general diet;(2)HC modle group (n-8)were fed with high-fat diet;(3)Chinese herbs(CH) treatment group(n=8)were fed with high-fat diet, and Resolving stasis eliminating phlegm granule was added from the end of the 12th-18th week. (4)Western medicine treatment control group (n=8)were fed with high-fat diet, Ticlopidine HYDROCHLORIDE combined with atorvastatin was added from the end of the 12th-18th week. Before and after treatment (at the end of the 12th and 18th week) liporotein(TC, TG, LDL, HDL) , P-selectin(ps) , Nitric oxide (NO) and Endothelinl (ETl)were measured. Animals were killed by the end of the 18th week, the samples of arteries and hearts were taken for morphologic analysis.Results (1) Before treatment, the level of TC, TG, LDLC and PS, ET-1 in h-c, CH and WM central group was markedly higher than those in the normal group(p<0. 05, p<0. 01) ;While the level of HDLC and NO was obviously lower (p<0. 05, p<0. 01) ; ㏕he CH gtoup had higher levels of plasma P-selectin(ps) and ET-1 compared with those of normal group(p<0.01);while lower levels of serum NO compared with those of normal group p<0.01); A direct positivecorrelalation was observed between plasma P-selectin and ET-1(r=) and a direct negative correlation between plasma P-selectin and NO levels (r=) ;(3) But no obvious differences appeared between CH group and WM control group (p>0. 05). After CH treatment TC, TG, LDL, PS and ET-1 significantly decreased(p<0. 05. p<0. 01), while the levels of HDL, NO were remarkably increased (p<0. 05, p<0. 01), comparing with the HC group. Conclusions The platelet activation, local inflammation reactin, the othelium unfunction which exists in and is closely related to the arterio-sclerotic lesions. And it can blocked by interventiog of resolving ctasis and eliminating thlegm.
