Juan Tong Tang Improve Hemorheology And The Experimental Study Of The Sedative Effect In Mice

Juan Tong decoction, a TCM formula for postherpetic neuralgia (PHN), which evolved from Moschus Pills in Pu Ji Ben Shi Fang, are composed of Scorpio, Lumbricus, Radix Aconiti, Flos Albiziae and Moschus, its actions are quickening blood, freeing network vessels, tranquilizing the mind and relieving pain; this dissertation reported its some pharmacological actions and discussed the relationship between pharmacological action and therapeutic effect. 1.Ameliorating hemorrheology parameters of blood-stasis-rat induced by adnephrinThree doses of Juan Tong decoction, 8.7g/kg (A), 2.9g/kg (B) and 1.0g/kg (C), were tested by intra-gastric administration. The results are: (1) A can degrade the whole blood viscosity, B offers weak effect and C has no effect; (2) A degrade the relative index of the whole blood's high and low shear, but B and C have no effect; (3) A can degrade the whole blood's high-shear-reduced-viscosity but can't degrade low-shear-reduced-viscosity, B and C have no effect; (4) A and B can degrade numerical value of haemat of normal rat and degrade akaryocyte-rigidity-index of blood-stasis-rat. (5) A and B can degrade the content of blood fibrinogen of normal rat. 2.1nhibiting the ambulation of miceJuan Tong decoction (3.0g/kg, 2.0g/kg, 1.0g/kg) can significantly inhibit the ambulation of mice, its effect is concerned of dosage but isn't positive correlation; its effect intensity is weaker but duration is longer than diazepam. 3.Extending the hours of sleep of mice controlled by pentobarbital sodium in threshold dosageJuan Tong decoction (2.0g/kg, intra-gastric administration) can extend the hours of sleep of mice controlled by pentobarbital sodium in threshold dose (50mg/kg, peritoneal injection) from 94.17?5.45min to 180.38?6.85min (PO.01), but provide no hypnosis effect when single application or pentobarbital sodium dose in 30mg/kg.4. Analyzing of relationship between pharmacological actions of Juan Tong decoction and its therapeutic effectIn western medicine system, the generation of PHN is concerned of nerve damage and dysimmunity; drugs for PHN mainly are: tricyclic antidepressant, acesodyne, NSAIDs, ect. In Juan Tong decoction, Moschus has nerve-protective effect; Scorpio, Radix Aconiti, Lumbricus and Moschus have analgesic effect; Scorpio inhibits immune and Lumbricus regulates immune; Scorpio, Lumbricus, Radix Aconiti and Moschus have anti-inflammatory effect; Flos Albiziae is an antidepressant. So, these pharmacological actions provide therapeutic effect.In Chinese medicine system, PHN is regarded as certain kind blood and network vessels stasis. Findings in this article indicated that Juan Tong decoction could ameliorate hemorrheology parameters, mitigate restlessness or dysphoria and relieve pain (reported in another article). These pharmacological actions can elucidate the therapeutic effect of Juan Tong decoction using under Chinese medicine theory.
