ObjectiveWe established the rats in diabetic nephropathy model. Through observing the effect of the TCM decoction TangShen HuaYu Jian (TSHYJ) on the blood glucose. plasma endothelin (ET), serum nitric oxide (NO), serum creatinine ( Scr), blood urea nitrogen ( BUN ) , kidney weight, index of kidney hypertrophy(kidney weight/body weight) on diabetic rats induced by injecting Streptozotoci ( STZ) into abdomen , to investigate the mechanism of the preventive and therapeutic effect of TSHYJ on the early diabetic nephropathy (DN) and provide an experimental basis for further research ,development and clinic medication.MethodsThe diabetic rats were induced by injecting Streptozotocin into their abdomen successfully, and they were randomly divided into five groups: normal control group (normal group) diabetic model group (model group) low dose of TSHYJ group (low dose group ) ; moderate dose of TSHYJ group (moderate dose group ) ; high dose of TSHYJ group (high dose group). A group had twelve rats. The decoction of TSHYJ was given to the three treated groups and the same dose of sodium chloride injection was given to the normal and model groups by stomach rearing once per day for six weeks. After these treatment killed all the rats to collect the blood samples, tested blood glucose, plasma endothelin (ET) , serum nitric oxide (NO), serum creatinine (Scr) , blood urea nitrogen ( BUN ), kidney weight and index of kidney hypertrophy. The kidney pathological changes of every group were observed under light microscope. All the results were analyzed with SPSS 10.0 for windows.Results1 Blood glucose There were no significant differences among model group and all treated groups in blood glucose before treatment (P>0.05) but the levels of blood glucose in all these groups were higher than that of the normal group (P < 0.01) . After treatment for 6 weeks, the level of glucose in the three treated groups were lower than that of model group (P < 0.05 or P < 0.01) .The moderate dose group had the best effect on lowering the level of blood glucose.2 BUN and Scr in serum (1)BUN: There were no distinct differences among all groups (P>0.05). (2)Scr: Scr of all treated group was distinctly lower than that of model group (P < 0.01) .3 ET and NO Plasma ET and serum NO of all treated and model groups distinctly increased, compared with the normal group( P < 0.05 or P < 0.01); Plasma ET and serum NO of all treated groups were significantly lower than that of themodel group (P < 0.01) , and moderate dose group had the best effect among them.4 Kidney weight and Index of kidney hypertrophy (1)Kidney weight: Kidney weight of modeh high and low dose groups were distinctly heavier than that of normal group (P < 0.01) moderate dose group had no change (P>0.05) . (2)Index of kidney hypertrophy: Index of kidney hypertrophy of the three treated and model groups were distinctly higher (P< 0.05 or P< 0.01) . TSHYJ could markedly decrease the index of kidney hypertrophy, compared with the model group ( P < 0.05 or P < 0.01 ) .5 The results of pathological sections under light microscope showed the curative effect of high and moderate dose groups were better than that of low dose groups.ConclusionTSHYJ had a favorable preventive and protective effect on the early DN rats, and the effect of moderate dose of TSHYJ group was best in all treated groups. The mechanism of its effects may be performed by lowering blood glucose , suppressing the kidney hypertrophy and reducing the level of ET and NO and improving renal function and the renal hemodynamic factors.
