ObjectiveThrough observing the effect of the Su-Jiang-Tang(SJT) on the blood glucose, blood pressure, body weight, kidney weight, index of kidney hypertrophy(kidney weight/body weight) , blood uria nitrogen and creatinine , urinary albomin excretion(UAE) , expression of transforming grown factor β1(TGF- β1) and accumulation of collagen Ⅳ (C- Ⅳ) in kidney tissue in diabetic rats induced by injecting STZ into abdomen ,to investigate the mechanism of the protective effect on the early diabetic nephropathy and provide an experimental basis for further developing and clinic medicating SJT.MethodsThe diabetic rats were induced by injecting Streptozotocin into their abdomen successfully, and they were randomly divided into five groups: diabetic model group " model group "(12 rats) , low dose of SJT group " low dose group " (10 rats) , high dose of SJT group "high dose group"(10 rats), Metformin group "drug Ⅰ group" (10 rats), Metformin and Losartan group "drug Ⅱ group" (10 rats);and other 10 normal rats were the normal control group "normal group ".The respective dose of drugs was given to the treated groups and the same dose of sodium chloride injection was given to the normal and model groups by stomach rearing once per day for eight weeks. After therapy, the body weight, the blood glucose, blood pressure, kidney weight > index of kidney hypertrophy , blood uria nitrogen and creatinine in serum, urinary albomin excretion(UAE), expression of transforming grown factor β1(TGF- β1) and accumulation of collagenⅣ(C-Ⅳ) in kidney tissue of rats of each group were tested. All the results were analyzed with SPSS.results1 Body weight There were no significant differences among all groups before treatment, P>0. 05. After treatment ,the body weight of the high dose group was distinctly heavier than that of the model group , drug Ⅰ group % drug Ⅱ group respectively, P<0.01or P<0.05.And the effect of the low dose group, drug Ⅰ group, drug Ⅱ group had no obvious difference before and after treatment, P>0.05.2 Blood glucose There were no significantly different among the blood glucose of all the treatment and model groups(P>0.05). The blood glucose of every treatment group was more lower than that of the model group, P<0.05. The blood glucose of the high dose group was more lower than that of the drug Ⅰ group, drug Ⅱ group respectively, P<0.01.The effect had obvious difference among all the treatment group before and after treatment , P < 0 . 0 5 .3 Blood pressure The blood pressure of the high dose group and drug Ⅱ group was more lower than that of the model group, P<0.05.4 Index of kidney hypertrophy SJT could markedly decrease the index of kidney hypertrophy, compared with the model group, marked by a distinct difference, P< 0.05. The effect between the high dose group and drug II group was no distinct difference,,5 BUN and Cr in serum There were no obvious differene among all the groups.6 UAE SJT and drug Ⅱ group could decrease the UAE markedly,compared with the model group7 TGF β1 and C-IV SJT could obviously down-regulate the expression of TGF β1 and decrease the accumulation of C-Ⅳ in the kidney tissue. The effect between the high dose group and drug Ⅱ group was no obvious differenceconclusionSJT has certain protective role on the rats with the early diabetic nephropathy to decrease the kidney hypertrophy and the urinary albomin excretion. The probable acting mechanism is to lower the blood glucose % down-regulate the expretion of TGFβ1 and reduce the accumulation of C-Ⅳ in kidney tissue.
