Purpose: In order to explain the mechanism of the MIGUKELI treating to osteoporosis from the gene expression level, we observed the effects of MIGUKELI to the osteoporosis rat due to Kidney-Deficiency and the osteoblast cultured in vitro.Method: Using the method of ovariectomy, we established the rat model of osteoporosis due to kidney-deficiency; and we also cultured the osteoblast in vitro. Using dual energy X-ray bone density instrument XR-36 model to detect the bone mineral density of the left femur and tibia of the rats; we also examined the expression of IGF- I mRNA gene in the bone tissue of osteoporosis rats due to Kidney-Deficiency and the osteoblast cultured in vitro with the method of RT-PCR; using the method of immunohistochemistry to research the expression of IGF- I in the bone tissue of osteoporosis rats due to Kidney-Deficiency; and measuring the activity of IGF- I in the serum of the osteoporosis rats due to Kidney-Deficiency; we identified the osteoblast cultured in vitro with the method of enzyme cytochemistry(ALP-Gomori) and evaluated the proliferation of the osteoblast after the action of serum contained tonic-kidney herbs.Result: In vivo part, we found MIGUKELI can increase the expression of IGF- I mRNA gene evidently, this kind of function is more excellent than the GUSHUKANG and nilestriol. Through the experiment, we also proved the IGF- I expression of the MIGUKELI group enganced obviously than the GUSHUKANG group and nilestriol group. Its IGF- I activity in the serum is better than the control group. In vitro part, the serum contained MIGUKELI herbs has the function of stimulating the proliferation of the osteoblast, Both the serum contained MIGUKELI herbs and the serum contained GUSHUKANG herbs ^ the serum- contained nilestriol can increase the IGF- I mRNA gene expression of the osteoblast in vitro, all of the functions have dose effect dependence.Conclusion: MIGUKELI may promote the bone formation process through affecting the system of Insulin growth factor- I . MIGUGELI can prevent and treat the osteoporosis.
