ForewordSquid ink is a kind of self - protective material when it encounter its natural enemy. It contains large amount of black coloring matter. In the early days of 1990s, Japanese researcher found that squid ink has the antitumor activity with high efficiency, then many researcher studied the its immunity about antitumor and the mechanism of it for the exploitage of squid ink on antitumor and immunopromoting activi-ty.There are many domestic research about antitumor mechanism of squid ink , mainly fasten on the immunomodulating activity of it. Our laboratory do a lot of work about the effects of its extracts on the specific and nonspecific immune function.The results of Lu changlong et al show that squid ink has the activity of stimulating and enhancing macrophage ; it can remarkably increase the levels of TNF - , IL - 1 and IFN - in the mouse serum; it also has .the promoting function on the killing activity of NK cell and LAK cell. It indicates that the antitumor effects of aquid ink is due to the enhancement of immune function, especially, the results of enhancement of cellular immunity.In recent years, many research show that there is a kind of new material , nitric oxide ( NO ) , which is a kind of small inorganic molecule. NO plays an important role in the process of activated macrophage killing the tumor cells. NO is a newfounded antitumor molecule, the induction of NO is the main mechanism of activated macrophage killing the tumor cells, a lot of cytokine can induce the macro-phage to synthesize amount of NO. it is showed that IFN - 7 has the obvious induction effects,it promotes macrophage to secrete NO.In a conclusion , squid ink and its extracts have the remarkable antitumor activity ,but it does not exhibit the direct cytotoxicity to the tumor cells, it hints that it exerts its antitumor effects indirectly via stimulating the body immune system. It is proved that squid ink and its extracts can modulate immune system via inducing the cytokines and effetor cell, at the same time ,we can not exclude the possibility of the produce of other effector molecule and cells, which regulate each other to carry out their antitumor effects, during its induction . then it is not clear till now that if there is some necessary relation between NO and the antitumor activity of squid ink. This article act as a part of this kind of research, based on the past researches , studing the effects of squid ink on the NO secretion level of celiac macrophage and its mechanism , expecting to do some help to the illustration of the antitumor mechanism of the squid ink.Material and methodTake 30 C57BL/6 mice, divide into 6 groups ,5 mice each group, among which the fifth group is the experimental group, in the experimental group each mouse is treated orally with 5% concentration squid ink 0. 5 ml each time ,one time per day, the control group are treated with NS by the same method as the experimental one. Assume the spleen cells and celiac macrophages of the mice at the zero day , second day ,forth day, sixth day , eighth day after treated orally with squid ink , use Griess method to determine the NO2- level and use ELISA method to determine the IFN - -y content. In addition, the spleen cells alone , adhesive cells and nonadhesive cells of the micetreated orally with squid ink the sixth day are cocultured with LPS or L - NMMMA. observe its NO2- producing level.Result1. effects of squid ink on NO yield level in mouse spleen cells using Grisee method ,we found that NO2- level in the culture ofmouse spleen cells which were treated orally with squid ink after 4 days began to rise (4.4 0. 74urnol/L) ,and got the highest level at the sixth day(9. 11 0. 88 mol/L) ,then began to descend (4. 1 0. 79 mol/L). the NO level in the culture of the cells of the group treated with squid ink orally on the forth day, the sixth day and the eighth day were much higher than the control group (2. 5 1. 0 mol/L) ( P <0.01)2. effects of squid ink on NO yield among the celiac macrophages of the mi...
